Objective:Explore the vascular endothelial cell injury inducing to MODS from the molecular and the overall two levels in SAP. Explore the protective effect of injury mechanisms and albumin on vascular endothelial cells injureded by trypsin. Methods:270SD male rats were randomly divided into6groups (n=45). Grouped as follows: Group A control group of sham operation group:laparotomy only gently flip the duodenum and pancreas, the abdomen was closed; Group B model of SAP Group:retrograde injection of5%sodium taurocholate into the pancreatic duct. Induced model of SAP; Group C octreotide group of Hospice Group:SAP group+octreotide; Group D ulinastatin groups:SAP group+octreotide+ulinastatin; Group E the albumin groups:SAP group+octreotide+albumin; Group F:SAP group+octreotide+ulinastatin+albumin. Groups point in time and then randomly divided into three groups, respectively,3h group,6h group,12h group,15in each treatment group animals after modeling30minutes subcutaneous injection of Sandostatin25μg/kg. In addition, ulinastatin10000U/kg;5%albumin10ml/kg; intravenously by rats. Each group of rats were sacrificed at different time points randomly femoral vein blood, and collect the tissue samples of pancreas, liver, kidney, lung and other organs. Measured in each group serum AMY, ALB, ALT, BUN, Crea and other indicators; above organs and tissues were morphological examination with reference to Schmidt’s score on the SAP group at each time point in pancreatic tissue of rats score. Flow cytometry to count off blood circulation vascular endothelial cells (CEC). Determination of vascular endothelial cell tight junction occludin protein expression using immunohistochemical staining.Results:5%sodium taurocholate into the biliopancreatic duct injection successfully induced severe acute pancreatitis complicated with MODS rat models. Serum AMY, ALT, BUN, Crea, the CEC count was significantly increased3h after the success of the modeling, is a rising trend, peaked12h, while ALB and endothelial cell tight junctions occludin protein expression gradually decreased with time. The albumin could antagonize the the pancreatin to closely to connect pairs the vascular endothelial the the damage of the protein, thereby protection the vascular endothelial cells, maintenance the the integrity of the of the vascular endothelial barrier, reduce the the the the occurrence of rate of of the complicated with MODS in the SAP. achieve the protection the the role of of distant organ.
