Part1Establishment of a young rat model with metabolic syndrome disease and the study of vascular pathological changes and immunoreactivity of VEGF、vWFObjective:To induce metabolic syndrome by giving high fat and high salt diet to detect the influence of high fat and high salt diet on W, AC, sugar metabolism index,lipid metabolism index,and the pathological change of vascular. Study the relationship between VEGF、vWF and the risk of cardiovascular disease in MS.Methods:Four-week-old SD rats were randomly divided into3groups, routine diet group (NC), high fat diet group (FC) and high fat+high salt diet group (FSC). The values of W, AC, BP, visceral fat weight, plasma lipids, OGTT test were measured4weeks after experiments. Immunohistochemistry were used to detect the expression of VEGF, vWF in the abdominal aorta, Use ELISA to detect serum VEGF, vWF.Results:l.The W, AC, visceral fat weight in FSC and FC group were significant higher than the control group (P<0.05or P<0.01).2. The blood TC, TG, HDL, LDL, FBG, FIN in FSC group were significant higher than that of the NC group(P<0.05or P<0.01).The blood LDL、AI、FBG、FIN、HOMA-IR in FC group were significant higher than that of the NC group(P<0.05or P<0.01)3. Hematoxylin-eosin staining shows:NC abdominal aortic endothelial cells of rat was flat single arrangement, morphological rules; The cells in FC have complete structure, arrangement; Some cells of FSC fall off, arrangement owe tidy, muscle layer of cells are arranged disorder, the nucleus polarity is poor, appear atypia. Not see clear foam cells or fatty streaks formation.4.VEGF and vWF immunohistochemical in FSC group express higher than NC group (P<0.01).The VEGF in FC group express higher than NC group (P<0.05).Both in serum and immunohistochemical express the same.5.Multivariatestepwise regression analysis suggests that AC(P=0.003), DBP (P=0.009),HOMA-IR(P=0.048),AI(P=0.008) is the independent risk factor of VEGF; HDL(P<0.001),TG(P=0.01)is the independent risk factor of vWF.Conclusion:1. High fat and salt diet for4weeks, the FSC group rats appeared obesity,high blood pressure, lipid disorders, sugar metabolism disorders, so the high fat and high salt induced young rat MS model success.2. Compared with known adult rats MS model,the vessels pathological changes lighter in young rats with MS,without any obvious atherosclerosis change.3The abdominal aorta and serum VEGF, vWF expression in FSC group higher than NC group,So there has vasofunctional disturbances long time before the appearance of a clear sclerosis of arterial congee. VEGF, vWF can indicate early vascular function monitoring.Part2:The relationship between VEGF vWF and the risk of cardiovascular disease in children with Metabolic SyndromeObjective:To study the relationship between serum activity of VEGF、vWF and the risk of cardiovascular disease in children with MS.Methods:30MS children,30simple obese children and30normal children(control group) were measured H,W,WC,HC,BP;Do carotid artery ultrasound in MS and simple obese children; to detect TC,TG,LDL,HDL,VEGF,vWF,FBG,FIN.And the data were analyzed statistically.Results:1.The H, W, BMI, WC, HC, WHR, BP, FBG, FIN, HOMA-IR, TC, TG, HDL, LDL, AI in MS group were significant higher than the control group(P<0.05or P<0.01).2.Beside H, TC,TG and vWF, other index in simple obesity group were significant higher than the control group(P<0.05or P<0.01).3Correlation analysis and multivariatestepwise regression analysis suggests that there is a positive correlation between serum VEGF and WC. HC、WHR、FBG、FIN、HOMA-IR、 R-CAIMT、L-CAIMT(P<0.05or P<0.01), HOMA-IR (P=0.005) is the independent risk factor; vWF and FIN、R-CAIMT、L-CAIMT has a positive correlation (P<0.05or P<0.01), vWF and HDL has a negative correlation(P<0.01),HDL is the independent risk factor.Conclusion:1.VEGF, vWF involved in the MS children of the physiological processes of cardiovascular disease, they can be used as a biomarker of the early diagnosis of cardiovascular risk.2.If MS children have glucose metabolism and lipid metabolism disorders, we should pay more attention to the risk for vascular injury.
