Expression And Correlation Research Of HIF-1α, INOS In High-risk HPV Positive Patients With Cervical Lesions

Objective:To detect the expression of hypoxia inducible factor-1a (HIF-1a) and inducible nitric oxide synthase (iNOS) in high-risk HPV positive patients with chronic cervicitis,cervical intraepithelial neoplasia and cervical cancer and find their effects and relations in the development of cervical lesions,investigate their clinical significance.Methods:Selected129cases cervical paraffin tissue samples including chronic cervicitis20cases,CIN Ⅰ grade30cases,CIN Ⅱ grade30cases,CIN Ⅲ grade23cases and cervical carcinoma26cases with high-risk HPV infection.HIF-la protein and iNOS protein were detected by immunohistochemical SP method.Results:(1)The positive expression rate of HIF-1a in chronic cervicitis group, CINⅠ grade group, CINII grade group, CINⅢ grade group, the cervical cancer group, respectively,0,10%,33.33%,47.82%,76.92%, the rise trends in five groups, compared to a statistically significant difference (P<0.05). The compares of the expression of HIF-1a in cervical cancer group and CIN group and control group, differences were statistically significant (all P<0.05), the expression of HIF-1a in CINⅢ grade group and CINⅡ grade group were statistically significant (P <0.05),compared with CINI grade group and control group. In CIN Ⅲ group and CIN Ⅱ group there was no significant difference (P>0.05). In CINⅠ grade group and control group there was no significant difference (P>0.05).(2)The positive expression rate of iNOS protein in chronic cervicitis group, CINI grade group, CINⅡ grade group, CINⅢ grade group, the cervical cancer group, respectively,5%,20%,26.67%,56.52%,84.62%, the rise trends in five groups, compared to a statistically significant difference (P<0.05). Expression in cervical cancer group and CIN group and control group, the differences were statistically significant (all P<0.05); The rate of iNOS protein positive expression in CINⅢ grade group was statistically higher than that of control group, CINⅠ grade group, CINⅡ grade group.(P<0.05). In CIN Ⅲ group and CIN Ⅱ group there was no significant difference (P>0.05).The compares of the expression of iNOS in CINⅡ grade group and control group, CINI grade group and control group, were all not statistically significant (p>0.05).(3) The correlation of HIF-1a and iNOS was positively correlated in high-risk HPV positive patients with cervical lesions by using Spearman correlation analysis (rs=0.869,P<0.005)Conclusion:HIF-1a and iNOS play collaborative role in promoting the development of high-risk pattern of cervical cancer HPV infectioned. Both express lifts is cervical cancerous process early behavior, which can help to predict the progress and outcome of cervical lesions.
