Surface enhanced Raman scattering technique as the identification of molecular species of the most powerful analysis tool, has the advantages of high sensitivity, strong specificity, good stability and detection of gentle reaction condition. At present, SERS technology has been used in the field of tumor pathology, although in its infancy, but has shown a good prospect and development space. With the progress of the testing equipment, laser technology and nano-preparation techniques, can be predicted that SERS will become the clinical pathology and laboratory diagnosis, and pathological study of a powerful means of detection and analysis tools, whether it is in the detection of cancer, or in the judgement of prognosis, monitoring efficacy and other aspects will play an important role.In this study, we applied diffent active basement, and preparated of the corresponding SERS immune probe, on colon cancer and nasopharyngeal carcinoma, blood samples of colon cancer for SERS immune detection, the main research contents are as follows:First of all, using Au Core-Ag Shell Nanoparticles for enhanced substrate, with4-MBA as SERS marker, we prepared CEA-SERS immune probe, which is used to detect CEA expression in colon cancer tissue and detected good SERS signals of probes in colon epithelial cells. Then we used the silver nanoparticles for enhanced substrate, preparation of crystal violet (CV) markers and4-Mercaptobenzoic Acid4-mercapto acid (4-MBA) labeled SERS probes, on colon cancer tissue for two-component immune detection. Next, we used4-MBA labeled CEA-SERS immune probe for quantitative detection of CEA in colon cancer blood. Finally, we used4-MBA marked LPM1-SERS immune probe for SERS immune detection on nasopharyngeal epithelial tissue, and achieve better results.Studies have shown that, the method of SERS immune detection has the advantages of simple, fast, and easy, and also for the biological samples two-component immune detection, which improve the efficiency of the analysis, you can get a variety of test results of objects, The method shortens the detection time, simplifies the analysis process, reduce the detection cost, and has higher application value.
