Chinese people have habit of cooking food in a pan over direct heat in hot oil or fat. This cook manner makes a lot of cook oil fumes, which is one of sources of aerial pollution. Many studies showed that cook oil fumes contribute to human cancer because of their ability for induced mutation. In this study, we liked to investigate the toxicity of concoctive oil fumes on SD rats’lung and to give some tactics to prevent cook s from their’harm.Forty-eight SD rats, which weight was (190±15)g, were divided into two groups random. Each one was averagely subdivided according to the times (1,15,30,60d) of exposure to cook oil fumes. Exposure device was designed by us. Exposure dose was adjusted to (43.82±11.58) mg/m3. Alveolar macrophages(AM), lactate dehydrogenase(LDH), alkaline phosphatase(AKP), acid phosphatase(ACP) in the bronchoalveolar lavage fluid(BALF) were detected at different time point. Windpipe and lung were put into10%neutral formaldehyde solution immediately, fixed, the ethanol grads dehydrated, the normal regulations paraffin wax embedded, cut into slices5-10pieces, HE dyeing, observed morphology variety with light microscope. Malondialhehyde(MDA) of the lung organization was measured with TBA colorimetry. Calorimetric test kit was used to measure total anti-oxidative capability (TOC) in the supernatant fluid of lung homogenate.At the first day, the number of AM in the exposure group was declined remarkably, compared with the control group[(2.76±0.67) X106/ml vs (14.92±1.26) X106/ml].With the exposure time going, the number of AM went up to the normal level[(4.93±0.83) X106/ml vs (4.87±0.84) X106/ml], but it was down remarkably at60d[(3.14±0.72) X106/ml vs (5.03±1.15) X106/ml]. Activities of LDH,ACP and AKP of the exposure group were increased to a certain extent, compared with the control group. It was remarkable in LDH. ACP and AKP only increased at the beginning. Morphology varieties of windpipe and lung showed that the cilia of bronchus epidermis fall of, lodging, adhere and the goblet cell increases, the lymphoid cell under mucous increases obviously, the lung appeared the lobular pneumonia (with lymphoid cell infiltrated), also amalgamation pneumonia. With time postponing; on the bronchus epidermis scale metaplasia, the mucous sheds of, the goblet cell increases obviously. The lung presents the interstitial pneumonia, the macrophages infiltrate around windpipe in lung. The TOC of the lung was obviously lower than control group(P<0.05), but the activity of MDA was obviously higher than control (P<0.05).We draw a conclusion that concoctive oil fumes do harm to AM and lung of SD rats. Cleanliness setting and preventing cook from polluters of concoctive oil fumes are necessary to our health.
