Objective: To study whether1,25-dihydroxy vitamin D3can protect diabeticretinopathy in rats and investigate the impact of1,25-dihydroxy vitamin D3on the levelof TGF-β1, VEGF expression in the retina of diabetic rats. Methods:35male SD rats,take10as the normal control group, inject Streptozotocin (STZ) into enterocoelia of therest rats,to establish diabetes model, Excluding3rats which are dead or not successfulmodel rats, the rest are randomly divided into two groups: diabetes group and1,25-(OH)2D3treatment group, Per group had11rats.1,25-(OH)2D3treatment group arefilled the stomach with1,25-(OH)2D3dissolved in peanut oil by0.03μ g/kg.d,diabetes group and normal control group are filled the stomach with equal peanut oil. AfterFeeding eight weeks, anesthetize and put the rats to death, reserve retinal tissue, observe theretina morphological changes with hematoxylin-eosin staining and detect TGF-β1, VEGFexpressions in the retinal tissue with the immunohistochemisty. Results: Compared with thenormal control group, TGF-β1, VEGF expression of diabetes group and1,25-(OH)2D3treatment group significantly increased (P<0.05). TGF-β1, VEGF expression of1,25-(OH)2D3treatment group are significantly less than that of diabetes group (P<0.05). The pathological observation: diabetes groups: retinal pigment epithelium layer cellsedema, photoreceptor cell layer and bipolar cell layer edema and thicken, bipolar cell layercells are arranged in disorder.1,25-(OH)2D3treatment group: The layer of cells arranged inbasic order, cells with mild edema, retinal thickness slightly thickened. Conclusions:1,25-(OH)2D3has some protective effect on diabetic retinopathy, its mechanism is that it maycan curb TGF-β1,VEGF protein expression in the retinal tissue.
