Objective: In order to understanding the fungal infections and Erg11gene mutationfrom AIDS patients, In this paper, the characteristics of fungal infection and therelationship between the target gene Erg11and azole antifungal agents isolated from AIDSpatients was investigated. Methods: The identification and susceptibility testing wereexecuted for stranis.The Erg11genes of c. albicans were amplified by polymerase chainreaction (PCR), they were all sequenced bilaterally, The sequencing results were analyzedby DNAman software and were compared with the GeneBank X13296, finally Mutationbases would be translated into amino acids to determine whether some missense mutationoccured. Meaningful missense mutation were done gene cloning. Results: Candidaalbicans was common pathogens of AIDS patients, with a high azole antifungal-resistancerate.Forty bases mutations were detected, which included27silent mutations and13missense mutations. We found that E266D and V488I were unconcerned in azoleantifungal-resistance of candida albicans, and one or no missense mutation was detected inCandida albicans resisted to one kind of azole-antifungal drugs while two or threemissense mutations present in strains resisted to two or three kind of azole-antifungaldrugs. Conclusion: Candida albicans was common pathogens of AIDS patients, with ahigh azole antifungal-resistance rate. The interaction of multiple mechanisms resulted inc.albicans resisted to azole antifungal drugs. All of missense mutation of ERG11gene,coordinated role of missense mutations, transformation of missense mutations fromheterozygote to homozygote, drug discharge enhance, and Biofilm formation may realatedto azole-resistance.
