Objective:Diabetes mellitus, often simply referred to as diabetes—is a group of metabolic diseases characterized by high blood sugar (glucose) level, that result from defects in insulin secretion, or action, or both. Then causing a sugar, fat and protein metabolism disorders and gradually causing the other organs injury and dysfunction. In recent years, as China’s social and economic conditions to improve the people’s living standard, the continuous improvement of dietary structure change, reduction, labor intensity, the crowd average life expectancy increased stress of extended and diabetes testing means of improvement and as countries around the world, the prevalence of diabetes in diabetes to rise gradually. The Chinese diabetic patients have already ranking second in the world, at the breakneck speed. So the study the pathogenesis of diabetes, disease characteristics, some prevention measures and treatment is very significant. Gynostemma Pentaphyllum contain a variety of composition, have a variety of pharmacological effects, according to the report that Gynostemma Pentaphyllum have antiphlogistic function and fall blood fat function, it may also have the function of fall blood sugar. This paper was used the pharmacological experimental animals of mice and internal drug analysis as the further proof for this role, and try to explain it’s nechanism of reducing blood sugar, and provide certain theoretical basis for he clinical application.Methods:This paper use the extract yield of Gynostemma Pentaphyllum as he complete index, design the orthogonal experimental method to optimize water extraction and alcohol precipitation condition for Gynostemma Pentaphyllum. And find out the optimum technological conditions for total extract, then use macroporous resin to separate total extract into different polarity parts, according to different polarity parts to determine the animal experiment groups. Improve the model making method of experimental diabetic mice, make sure the injecting dose of STZ (chain with urea bacteria grain) which will be the best dose for diabetic mice model; Study the different polarity parts’s impact of Gynostemma Pentaphyllum on high blood sugar in mice (GLU), total cholesterol (TC), triglycerides (TG), superoxide dismutase (SOD), malondialdehyde (MDA) and free fatty acid (NEFA), through the various biochemical index level to determine which is the most effective part of Gynostemma Pentaphyllum on reducing blood sugar. Then use high performance liquid chromatography (HPLC) to determine it’s constitution of the effective part.Results:The optimal water extraction process was added10times water for3times and1.5hours for each time. The best alcohol precipitation technique was as follows:60%of precipitation concentration, then keeping depositing for24h. In our diabete mice models the most optimum dose is160mg-kg-1STZ for injection at one-time. Through the animals pharmacological experimental, we concludes that compared with the model group mice, Gynostemma Pentaphyllum70%alcohol part has very significant difference in reducing high blood glucose GLU and NEFA levels.(P<0.01), has significantly difference in increasing SOD level (P<0.01) and has significantly difference in reducing TC and MDA levels (P<0.05), water extraction part has significant difference in reducing GLU, TC and MDA (P<0.05)levels, however,30%alcohol part has no significantly difference compared with model group. Use HPLC to determine, there were three components may have major function on efficacy.Conclusion:(1) In order to provide the theoretical basis for Gynostemma Pentaphyllum is production. We use the orthogonal experimental to optimize the water extraction and alcohol sink technology.(2) Using the STZ method to establish diabetes model is one of the most applied method, Our experimental use one-time intraperitoneal inject of STZ at140,160,180mgkg-1three kinds of doses to analysis the blood sugar concentration and the stability of the model, This study in order to provided theory support for diabetes model.(3) When we screening the effective parts from the Gynostemma Pentaphyllum, we found that the total extractings from gynostemma pentaphyllum and the component I and III from the Gynostemma Pentaphyllum can different degree reducing the concentrations of blood sugar, component III has the more stronger effective than component I, however component II do not have such function. Except this the component III can also reduce of the high blood fat level, which diabetes caused, however the components I and II have the less ability in this function, This study provide the solid foundation for the further development of Gynostemma Pentaphyllum.(4) Analysis the active ingredients, we found that there may three kinds of components, further analysis from the HPLC data, found they may be flavonoids or saponins, this two types of material both have the pharmacodynamic effect, if they are saponins, may be because saponins has antiphlogistic effect, this imply that the inflammation has some thing to do with the diabetic.
