Aeschynanthus moningeriae is the species of Aeschynanthus genus of the family Gesneriaceae. The genus has about80species, and distributed throughout tropical and subtropical areas. There are24species in china, Introducing places such as south, and southwest. It is used in treatment inflammation, wheezing cough, furunculosis, rheumatism, bones injuries, empyrosis, snakebite and gynecological diseases. This paper took Aeschynanthus moningeriae as the study object, analysis from two aspects as following, provides the scientific basis for this plant resources’ development.1, The powdered Aeschynanthus moningeriae were extracted with70%methanol under room temperature to give a crude extract. After concentration, the residue was suspended in water and then partitioned with ethyl acetate and n-butanol successively to afford EtOAc fraction, n-C4H9OH fraction and aqueous residue. Application Diaion HP-20, Toyopearl HW-40, Sephadex LH-20, MCI-gel CHP-20, silica gel, RP-18isolation fillers and isolation techniques, fifteen compounds were isolated from EtOAc fraction, n-C4H9OH fraction. The structures of twelve compounds were identified on the basis of physiochemical properties and spectral methods(1HNMR,13CNMR, DEPT,1H-1H COSY, HSQC, HMBC, HR-MS).The twelve compounds were elucidated as:5,7-dihydroxy-8-C-P-D-glucosyl-chromone (â… ),5-hydroxyl-7,4’-dimethoxyl-flavone(â…¡),5-hydroxyl-6,4’-dimethoxyl-7-O-[a-L-rhamnosyl-(1â†'6)]-β-D-glucopyranosyl-flavone(â…¢),5,7,3’,4’-tetrahydroxy-6-C-P-D-glucosyl-dihydrofl avonol(IV),5,7,3’,4’-tetrahydroxy-6-C-β-D-glucosyl-flavonol(â…¤),5,7-dihydroxy-6-C-β-D-glucosyl-flavonones (â…¥),2,3,19,24-tetrahydroxy-urs-12-en-28-oic acid (â…¦), daucosterol(â…§),2,3,19,24-tetrahydroxy-urs-12-en-28-oic acid ester-28-O-β-D-gucoside (â…¨), stigmasterol(â…©), P-sitosterol(XI),3-0-β-D-glucosyl-stigmasterol(â…©â…¡). Compound I is a new compound, other compounds were isolated from this plant for the first time.2, The crude extract of70%EtOH(70%E-CE) was obtained by extracting the powder of Aeschynanthus moningeriae with a solution of70%ethanol. Then,70%E-CE dissolved in distilled water was fractionated with petroleum ether (PE), and ethyl acetate (EtOAc), respectively, affording three fractions of PE-EF, EtOAc-EF, and W-EF. Then, the W-EF was added to Diaion HP-20column, and the resin was eluted by distilled water,10%,30%,50%,70%methanol, respectively. The content of total flavonoids (TF) in different extracts and enrichment fractions was determined by the NaNO2-Al(NO3)3method, and the content of total polyphenols (TP) was determined by Folin-Ciocalteu method. Second, the antioxidant activities of different extracts and enrichment fraction were investigated by DPPH and FRAP method. In addition, the relativity among the content of TF, TP and antioxidant activity was also investigated. As result, the contents of TF and TP in CE’s fractions decreased in the following order:W-EF> EtOAc-EF> PE-EF.The content of TF and TP of enrichment fractions decreased in the following order:30%MeOH-EF>50%MeOH-EF>10%MeOH-EF>70%MeOH-EF. The TF content is higher than TP content in the same sample, and the contents of TF and TP in enrichment fractions were higher than that of CE’s fractions. The DPPH free-radical scavenging activities decreased in the following order:W-EF> EtOAc-EF> PE-EF;30%MeOH-EF>50%MeOH-EF>10%MeOH-EF>70%MeOH-EF. The total antioxidant activities decreased order is the same with DPPH free-radical scavenging activities.The TF, TP content and outcomes of two kinds of antioxidant activity methods had very significant relativity. It indicates that flavonoids and phenols are the main components responsible for antioxidant behavior of Aeschynanthus moningeriae, and there was very high correlation between the measuring results of these two methods, so they were all available for determination of antioxidant activity of Aeschynanthus moningeriae.
