Objective: To detect the differential expression of Notch1in the GT ofhypopspadiac male fetal rats induced by maternal exposure to DBP and normalcontrol fetal rats in order to further explore the mechanism of hypospadias.Methods: Twenty pregnant SD rats were randomly divided into two groupsacting as experiment group and control group, which were given DBP or soybean oilby gastric intubation at the dose of800mg/kg respectively from gestation day (GD)14to GD18. On GD19, birth weight (BW) and anogenital distance (AGD) were recorded,hypospadias incidence was calculated and GT of fetal rats were collected. Theexpression of Notch1was analyzed by western blotting and immunohistochemistry.Results: The incidence of hypospadias was42.9%. The BW of fetal male ratswas significantly lower in the experiment group (4.40±0.30, g) than that in thecontrol group (6.11±0.40, g)(P <0.05), and the AGD was shorter in the experimentgroup (2.17±0.18, mm) than the control (3.28±0.16, mm)(P <0.05). Notch1expressed both in the experiment group and the control group, and was mainlylocated in the cytoplasm of epithelium. The expression level of Notch1wassignificantly lower (P <0.05) in the experimental group than in the control group atGD19.Conclusion: DBP has obvious toxicity to fetal rats. DBP can reduce the notch1 expression in GT, and it could possibly affect cell proliferation, apoptosis andepithelial-to-mesenchymal Transition (EMT), and then result in the occurrence ofhypospadias. But its function needs in-depth study. Through hypopspadiac fetal ratsmodels, we further study the mechanism of hypospadias and the results could provideus some clues to prevent the reproductive toxicity of DBP.
