ObjectiveTo detect CD137L expression on human non-small cell lung cancer(NSCLC) celllines A549, H460, A2, SK-MES-1, SPC-A1and to investigate its effect on biologicalbehaviors in them.Methods1. The expression of human CD137L was detected by RT-PCR,FACS on the NSCLCcell lines including A549, H460, A2, SK-MES-1, SPC-A1;CD137L proteinexpression on A2was identified by western blot.2. A2was cultured on the dishes immobiled CD137-FC or IgG1-FC or PBS control.The cell proliferation was detected by CCK8at24h,48h,72h.3. The apotosis of A2cultured on the dishes immobiled by CD137-FC or IgG1-FC orPBS control was detected by Alexa Fluor488Annexin V/PI at24h.4. The expression of PD-L1on A2cultured on the dishes immobiled by CD137-FCor IgG1-FC was evaluated by FACS at48h.Results1. CD137LmRNA and CD137L protein are expressed on non-small cell lung cancercell line A549,H460,A2,SK-MES-1,SPC-A1at different levels.2. The A2cell proliferation of CD137-FC group was significantly increasedcompared with IgG1-FC and PBS group at24h,48h,72h(P<0.05).There was nostatistical significance between IgG1-FC group and PBS control group(P>0.05).3. The A2cell apoptosis of CD137-FC,IgG1-FC or PBS group has no significance(P>0.05).4. The expression of PD-L1on A2can be up-regulated by CD137-FC comparedwith IgG1-FC. ConclusionCD137L was expressed on several kinds of non-small cell lung cancer cell lines.CD137L mediated reverse signal can promote lung cancer cell proliferation andup-regulate the expression of PD-L1,while no influence on cells apoptosis was found.CD137L may play a role in ocurr and development of non small cell lung cancer.
