| Objective:Study the theoretical basis of Traditional Chinese Medicine treatment on joint diseases.Observe the curative effect of GuQing granules on the adjuvant arthritis rats,and detect if GuQing granules could influence the expression of acid-sensing ion channel3(ASIC3) in articular cartilage.Methods:1.Theoretical research:Through literature collection, to explore the genesis of the theory that "kidney being in charge of bone" and the Traditional and Modern medicine’s understanding of it. Review the research on the mechanism of tonifying kidney herbs that protect the articular cartilage.2.Experimental research:1)Fifty SD Rats were randomly divided into five groups:blank control group, adjuvant arthritis model group, positive control group (aspirin50mg/kg), therapeutical dose of GuQing granules group (8.4g crude drug/kg), high dose of GuQing granules group (16.8g crude drug/kg). Animals of the blank control group were injected intradermally into their left toes with sterile water, the rest were injected intradermally with Complete Freund’s Adjuvant (CFA) to induce adjuvant arthritis model.10days after proinflammatory, animals of each group were administrate with corresponding drugs for10days.2)Record the weight, volume of right foot and arthritis scores every3day. After execute the animals, ankle joints were taken out for observing the pathological changes by optical microscope, knee cartilages were taken out for analyzing ASIC3mRNA and protein expression by RT-PCR and Western Blotting.Results:1."kidney being in charge of bone" is one of the core theory of Traditional Chinese Medicine.Tonifying Kidney can promote the growth of bone and cartilage so as to make them strong.It is one of the principles on treating bone and joint diseases. Traditional Chinese Medicine consider that the growth and repair of bone depends on the promoting of kidney essence and the nutrition it provided. The destruction of bone is ascribed to the deficiency of kidney essence that cannot maintain bone growth and metabolism. Modern medicine’s understanding of "kidney being charge of the bone" is kidney secrete various hormones and cytokines which act on the physiology and pathology of bone. Modern pharmacological studies have shown that:tonifying kidney herbs treat joint diseases and protect cartilage through reducing the cartilage damage, delay cartilage degeneration and to promote both differentiation and repair work.the mechanism includes:(1) inhibit the expression of cytokines which promoting decomposition;(2) prevent free radicals formation, inhibit the activity of NOS, reducing NO formation;(3) the regulation of articular cartilage cell’s apoptosis;(4) effect on articular cartilage cell proliferation;(5) the role in the matrix of articular cartilage;(6) increase the expression of anabolic cytokine,etc.2.Animals in model group appeared a reducing on cibation, slowly in weight gain, some of rats even lose weight, they aslo get an seirous paw swelling.Compare with the model group, signs of rats in treatment group were obviously better. The volume of foot and arthritis scores have significant difference with modern group. Pathology and molecular biology experiments showed that articular cartilages of rats in model group were severely damaged, ASIC3mRNA and protein expression increased in knee cartilage. GuQing granules can significantly inhibit the paw swelling and the infiltration of inflammatory cell in rats ankle, significantly inhibit ASIC3mRNA and protein expression in knee cartilage, there was no significant difference compared with the positive control drug aspirin.Conclusion:"kidney being in charge of bone" is the theoretical basis of Traditional Chinese Medicine treating on joint diseases.GuQing granules can reduce the inflammation of adjuvant arthritis rats, reduce the cartilage damage and down-regulate the expression of ASIC3. |
