Aim To investigate the effects of over-expressed integrin-linked kinase on human umbilical vein endothelial cells(HUVECs).Methods HUVECs were digested by trypsin and cultured in vitro. Then,HUVECs were randomly divided into GFP (Ad-GFP) and integrin-linked kinase (Ad-ILK) virus transfection group. ILK was over-expressed in HUVECs by Ad-ILK transfection.Cells proliferation was measured by MTT, flow cytometry and EDU assay. The apoptosis of HUVECs were detected by flow cytometry under hypoxic conditions.Western blotting was used to assess expression and phosphorylation of Akt and eNOS.Results MTT assay, flow cytometry cycle, EDU assay all showed that cells proliferation were significantly higher in ILK over-expressed group than in the control group (P<0.01or P<0.05). Flow cytometry showed that the apoptosis rate was significantly reduced in the ILK over-expressed group (P<0.05). The phosphorylation of Akt and eNOS levels were also significantly higher than that in control group(P<0.05),but Akt and eNOS expression levels were no difference between the two groups.Conclusion Over-expression of ILK can promote HUVECs proliferation and prevent against apoptosis via actvation of Akt and eNOS.
