Objective: Schizophrenia (Sz) is a common spiritual activitiesincompatible with the environment as the main feature of mental illness, seriously affect the health and quality of life of the patient’s body to bring aheavy burden on the mental and economic, social and family. In particular, forensic or compulsory medical measures, because there is no objectiveexperimental basis, often resulting in a very awkward situation. Sz its incidencein the world’s different cultures, geographic area, are about1%, but there is anincreasing trend. At present, the diagnosis of schizophrenia is mainly based onclinical symptoms, with the development of molecular biology, proteomics andgenomics has become a new way of mental illness. In which thesurface-enhanced laser desorption ionization time of flight mass spectrometry(SELDI-TOF-MS) technology has achieved significant results in the screeningof protein biomarkers with schizophrenia. Group experiments on differentmental illnesses, screening of differentially expressed peptides and MicroRNAsin order to provide an objective basis for diagnosis, to explore the methods ofidentification of the mentally ill of no insight led him to study the etiology andpathogenesis. Methods: The subjects are divided into six groups of145casesincluding33cases of schizophrenia, anxiety disorders in30cases,14cases ofdepression,11cases of organic mental disorders, schizophrenia immediate family members of36cases,21cases of healthy people. Usingsurface-enhanced laser desorption ionization time of flight mass spectrometry(SELDI-TOF-MS) and weak cation chip (the Ciphergen ProtienChip CM10)collected serum peptide mass fingerprinting, Biomaker Wizard software filterout the differences in peptide group. With statistically significant protein peaksprotein database (Swiss protein database at http//us.expasy.org/tools/tagident.html) will filter out the query, select from the possible and availableELISA kit for serum peptides identified. The results by single factor analysis ofvariance and independent sample test statistics with significant differences inserum peptides. Select no insight schizophrenia group, the treatment ofschizophrenia remission group, serum samples of healthy persons using theAffymetrix high-throughput microarray screening of MicroRNAs differentiallyexpressed. SAM software, unsupervised clustering analysis software initiallyidentified differentially expressed features MicroRNAs synthetic characteristicsof MicroRNA primer preliminary validation experiment.Results:1、Screening of15differentially expressed serum peptide (P <0.01)in six groups of subjects the Chinese Communists, with molecular weights of2058.06Da,2159.68Da,2651.54Da,2774.36Da,2912.61Da,3116.9Da,3168.95Da,3407.71Da,4050.38Da,4100.61Da,5642.83Da,5913.05Da,9186.42Da,2093.81Da,2221.92Da.'. ELISA assay that neuropeptide2inthe schizophrenia group in serum were significantly higher, its average contentand standard deviation of11.28±3.83, and content in the other group were 7.57±2.38,6.68±1.97,7.63±0.33,6.21±6.576.82±4.83. Measuredusing analysis of variance p=9.74E-06, the difference was statisticallysignificant meaning.3. No self-knowledge force of schizophrenia and normalcan be compared to a screening of the expression is raised trend microRNA(hsa-miR-1281_st, Fold Change frequency1.5); Two microRNAs showed adownward trend (of hsa-miR-2861_st, Fold Change≤0.64193,hsa-miR-638_st, Fold Change≤0.51624). No self-knowledge force ofschizophrenia group compared with schizophrenia in remission group werescreened out two downregulated microRNAs. They were hsa-miR-320b_st(Fold Change than0.43678), hsa-miR-320a_st (Fold Change than0.25056).Schizophrenia remission group and the normal control group screened adownregulation of microRNA (hsa-miR-1469_st, the expression of a multipleof0.6446).Conclusion:1. Mental illness in patients with a group of abnormalexpression of the polypeptide molecules, and schizophrenia, anxiety disorders, depression, trauma due to mental illness has its own expression of the abnormalcharacteristics of peptides.2. Anxiety disorders in patients with abnormalexpression of peptides in patients with schizophrenia is basically the same andshowed a parallel state, only the abnormal low level of prompts from themolecular pathology of anxiety disorders may be light schizophrenia3.Neuropeptide2may be associated with symptoms of mental illness.4.hsa-miR-1281_st in the serum of patients with schizophrenia characteristic high expression of hsa-miR-320b_st and hsa-miR-320a_st low expression maybe associated with schizophrenia self-control, the differential diagnosis ofschizophrenia, insightDiscussion of judgment and pathogenesis is important.
