Objective:To study the relationship between sperm DNA damage and intrauterine insemination (IUI) pregnancy rate. To analyze the optimization effect of PureSperm(silanized silica suspensions) density-gradient centrifugation on semen. Methods:46semen samples were obtained from36males who came to the reproductive center of the affilicated hospital of Luzhou medical college for IUI from October2011to March2012. Each semen sample was processing PureSperm density-gradient centrifugation, routine semen analysis, Diff-Quick staining and AOT before IUI. Results:1. After PureSperm density-gradient centrifugation sperm motility rate and normal sperm morphology rate were increased significantly (P<0.01), but sperm density was increased slightly with no significant difference(P>0.05).2. Sperm DFI was reduced obviously after PureSperm density-gradient centrifugation (P<0.01).3. There were negative correlation between sperm DFI and sperm density (P <0.05) and significant negative correlation between sperm DFI and sperm motility rate or normal morphology rate (P<0.01).4. Sperm DFI of IUI pregnancy group (10%) was lower than that of non-pregnant group (20.22%)(P<0.01).5. Semen volume of pregnancy and non-pregnant group had no significant difference (P>0.05), sperm density and sperm motility rate of pregnancy group were higher than that of non-pregnant group (P<0.05), normal sperm morphology rate of pregnancy group(25.33%) was significant higher than that of non-pregnant group(14.95%)(P<0.01). Conclusion: PureSperm density-gradient centrifugation can greatly optimize sperm DNA integrity. Sperm DFI can be used to select couples for IUI.
