Objective: To investigate the mechanism and clinicalsignificances of baicalein inhibiting the proliferation and apoptosis, bydetecting the expression of Livin and Ki67proteins in human breast cancer cellline MCF-7cultured in vitro. Methods:1. Human breast cancer cell linesMCF-7were treated with various baicalein concentration (5.0μmol/L,10.0μmol/L,20.0μmol/L,40.0μmol/L,80.0μmol/L and160.0μmol/L) atdifferent action time (24h,48h,72h) for measuring cell proliferation by WaterSoluble Tetrazolium-8(WST-8) colourimetry.2. The apoptosis of human breastcancer cell line MCF-7was detected with different baicalein concentration(5.0μmol/L,10.0μmol/L,20.0μmol/L,40.0μmol/L,80.0μmol/Land160.0μmol/L)in different action time (24h,48h,72h) using Annexin V-FITC/PI by FlowCytometry.3. The expression of Livin and Ki67protein were detected inhuman breast cancer cell line MCF-7by Immunocytochemistry (ICC), andMCF-7cells were treated with different baicalein concentration (5.0μmol/L,10.0μmol/L,20.0μmol/L,40.0μmol/L,80.0μmol/L and160.0μmol/L) atdifferent action time (24h,48h,72h). Results:1. The growth inhibitionratio(GIR) of human breast cancer cell line MCF-7was increased after beingtreated with different concentration of baicalein (5.0μmol/L,10.0μmol/L,20.0μmol/L,40.0μmol/L,80.0μmol/L,160.0μmol/L).There was a significantdifference among each group at different action time with the same concentration(P<0.05); There was a significant difference among each groupwith different concentration at the same action time(P<0.05), and the inhibitoryeffect of baicalein on the proliferation of MCF-7cells showed a time-dosedependent manner.2. There was a significant difference among each group ofthe apoptosis effect at different action time (24h,48h,72h) with the sameconcentration; there was a significant difference among each group withdifferent concentration(5.0μmol/L、10.0μmol/L、20.0μmol/L、40.0μmol/L、80.0μmol/L、160.0μmol/L) at different action time. Compared with the controlgroup and the concentration group, there was a significant difference(P<0.05).There was a significant difference among each group with differentconcentration. The effect of apoptosis of human breast cancer cell line MCF-7treated by baicalein showed time-dose-dependent property.3. There was asignificant difference among each group of the expression of Livin protein withthe same concentration at different time(24h,48h,72h)(P<0.05). There was asignificant difference among each group at the same time with differentconcentration (5.0μmol/L、10.0μmol/L、20.0μmol/L、40.0μmol/L、80.0μmol/L、160.0μmol/L)(P <0.05), compared with the control group and the concentrationgroup, which was a significant difference of the expression of Livin protein.The expression of Ki67protein was significantly different among each groupwith the same concentration at different time(24h,48h,72h)(P<0.05), except thatthere was no significant difference between24h and48h in the control group(P>0.05). There was a significant difference among each group at the same timewith different concentration(P<0.05), except that there was no significant difference between the control group and the concentration group of5.0μmol/Lat24h(P>0.05).4. There was a positive correlation between Livin andKi67(r24h=0.604, r48h=0.731, r72h=0.98, P<0.05). Conclusions:1. Theproliferation of human breast cancer line MCF-7may be inhibited by baicaleinin a time-dose dependent manner.2. Baicalein may promote the apoptosis ofhuman breast cancer line MCF-7related to time-dose dependency.3. There maybe a positive correlation between Livin and Ki67.4. Baicalein may inhibit theexpression of Livin and Ki67proteins of human breast cancer line MCF-7, andit is supposed that baicalein induces the apoptosis of human breast cancer lineMCF-7by inhibiting the expression of Livin and Ki67, and it may be provide apossible clinical application for treatment of breast cancer.
