Objective: To dectect the expression levels of miR-155and miR-142-5p inperipheral blood mononuclear cells(PBMCs) from patients with atopicdermatitis(AD) and provide a theoretical basis for further researches of its role inthe development and progression of AD. Materials and Methods:8patientswith AD and6cases of normal individuals were selected. PBMCs were separatedby human lymphocyte isolation liquid, miRNAs in PBMCs were extracted andseparated by miRNA extraction and separation kit. The expression levels ofmiR-155and miR-142-5p in PBMCs of AD patients and normal individuals weredetected by using real-time quantitative PCR. U6snRNA was used as endogenouscontrol, the relative expression levels of miR-155and miR-142-5p in PBMCs ofAD patients were calculated by2-ΔΔCTmethod and the correlation between therelative expression levels of miRNAs and the SCORAD of AD were analyzed.All datas was analyzed by SPSS17.0, α=0.05was regarded as the statisticalcriterion. Results:1. miR-155and miR-142-5p in PBMCs of AD patients can bedetected.2. Compared with normal control group, the relative expression levelsof miR-155and miR-142-5p in PBMCs of AD patients were up-regulated (P<0.05).3. There was correlative tendency between the relative expression ofmiR-155and the SCORAD(r=0.685, P=0.056), but no correlations between therelative expression of miR-142-5p and the SCORAD(r=0.238, p=0.497).Conclusion:1.There are miR-155and miR-142-5p in human PBMCs, which canbe detected conveniently by real-time fluorescent quantitative PCR.2.miRNA-155and miRNA-142-5p in the PBMCs of AD patients are up-regulatedand may play a certain role in the occurrence and development process of AD.3. There was correlative tendency between the relative expression of miR-155and the SCORAD, but no correlations between the relative expression ofmiR-142-5p and the SCORAD.
