Objective: To investigate the expression of FBXW7,Aurora-Aand P53genes in gastric carcinoma and its correlation with clinicopathologicfeatures of gastric carcinoma and to explore the role of the development of thevarious indicators in gastric cancer. Methods: The FBXW7,Aurora-A and P53gene were detected by HE staining and immunohistochemical S-Pmethods in gastric carcinoma tissue and normal gastric tissue,observe their individual and joint expression of the situation, and the relation ofexpression of FBXW7,Aurora-A and P53genes with the development,progression,metastasis and prognosis of gastric carcinoma tissue.1.The surgicalresection and paraffin blocks of gastric cancer confirmed by pathologyspecimens of60cases and20cases of normal gastric mucosa archive paraffinblocks were obtained from the Department of Pathology of the Third People’sHospital of Mianyang City, September2007to September2011.2.Theinclusion criteria:①All the patients were not given chemotherapy andradiotherapy before preoperation, and all the pathological specimens wereconfirmed as gastric cancer after postoperation.②All the specimens were fixedby10﹪formalin, embedded in paraffin,5μm serial sections.③20cases ofnormal gastric mucosa were all taken from a normal gastroscopy specimens.The exclusion criteria:①All the patients were given chemotherapy andradiotherapy before preoperation, and all the pathological specimens wereconfirm as gastric cancer after postoperation.②All the normal gastric mucosawere confirmed gastric cancer or suspected gastric cancer by pathology.3.Usingimmunohistochemical SP method, DAB color. Anti-FBXW7,P53and Aurora-Amonoclonal antibody to compare pathological observation records FBXW7,P53and Aurora-A expression in gastric cancer and normal gastric mucosa tissuesamples.4.The statistical date was analyzed by statistical sofeware13.0SPSS,FBXW7, P53and Aurora-A expression in gastric cancer with χ~2test, p< 0.05was considered to be significant. Results:1.The expression ofFBXW7,Aurora-A and P53genes in gastric carcinoma tissues,there existedsignificantly itatstiscal difference comparing with normal tissues(P<0.05).2.The expression of FBXW7, Aurora-A and P53genes in gastric carcinomatissues had no obvious relation with the patient’s age,gender,differentiationdegree,TNM stage and lymph node metastasis(P>0.05)and carcinomadiameter>5cm,distant metastasis of gastric cancer patients with FBXW7genepositive expression rate was significantly higher than that of gastric cancerpatients without distant metastasis,the difference was significant(P<0.05),carcinoma diameter>5cm,distant metastasis of gastric cancer patients withP53gene positive expression rate was significantly higher than that of gastriccancer patients without distant metastasis,the difference was significan(tP<0.05及P<0.01), carcinoma diameter>5cm,distant metastasis of gastric cancerpatients with Aurora-A gene positive expression rate was significantly higherthan that of gastric cancer patients without distant metastasis,the differencewas significant(P<0.05).3.The expression of FBXW7gene was negativelycorrelated with the expression of P53gene(P<0.05, r=-0.443),the expressionof Aurora-A gene was more significantly related to the expression of P53gene(P<0.05, r=0.408),but the expression of FBXW7gene had no significantrelation with the expression of Aurora-A gene(P>0.05). Conclusion:1The lowexpression of FBXW7protein and the overpression of P53,Aurora-A arerelated to the progression of gastric cancer.2The low expression of FBXW7protein suggest that FBXW7plays an important defensive role in gastriccarcinogenesis.the overexpression of P53and Aurora-A prompted the invasionability of gastric cancer,the high degree of malignancy.3.Combined detection ofFBXW7、Aurora-A and P53protein in gastric cancer may reflect some of thebiological behavior of gastric cancer,assessment of disease,development ofrational treatment program for clinical.
