| Objective:To detect the minimum inhibitory concentration (MIC) of Shuang-huang-lian, Xi-yan-ping and da-suan-su for Staphylococcus aureus ATCC29213. then the mutant selection window (MSW) will be measured for three Traditional Chinese Medicine injections combined with levofloxacin, investigated which Traditional Chinese Medicine injections can narrow the levofloxacin MSW, reducing its SI. The best one be used to investigate the effect of the drug combination with levofloxacin on the mutant selection window (MSW) in vitro to Staphylococcus aureus through establishment of the rabbit tissue cage infection model to study traditional chinese medicine injectionsand levofloxacin in combination can reduce the resistance of Staphylococcus aureus to levofloxacin in the body. In order to reduce the clinical resistance to fluoroquinolones provide a new treatment strategies. Methods:MSWs of shuang-huan-lian, xi-yan-ping and da-suan-su injects against staphylococcus aureus were tested by using the agar doubling dilution method and caculat the SIs. Then traditional chinese medicine injectionsand levofloxacin use of traditional chinese medicine injectionsinvestigated more than narrow the levofloxacin MSW to reduce its the SI and filter out the good antibacterial effects and can reduce Staphylococcus aureus of levofloxacin resistance in traditional chinese medicine injections for further in vivo studies, in vivo testing through the establishment of rabbit cage Staphylococcus aureus ATCC29213infection model, through the marginal ear vein injection,1/d, continuous administration of5days. The joint group to give the injection of levofloxacin the8mg/kg then give Shuang-huan-glian powder for injection92mg/kg treatment. The left group given the the8mg/kg levofloxacin treatment, each of the positive control group given levofloxacin8mg/kg then given cefazolin26mg/kg treatment, the negative control group was injected with the corresponding amount of saline. The joint group with the left group compared the occurrence of resistance and to restore the MIC of the growth of bacteria, to examine whether traditional chinese medicine injectionsin the body to reduce the resistance of Staphylococcus aureus to levofloxacin. Results:The MSW of Shuang-huang-lian is1.905～14.737mg/mL, the SI is7.736; the MSW of garlic injection is MSW5.960～38.500μg/mL, the SI is6.460; the MSW of Xi-yan-ping can’t be measured. Shuang-huang-lian narrow MSW of levofloxacin against Staphylococcus aureus ATCC292132.579-fold, and garlic injection of the SI to levofloxacin and MSW almost no effect. In vivo experiments show that the levofloxacin on Shuang-huang-lian group resistance, was20%, to restore the growth of bacteria, the MIC of0.728±0.285, single rate of100%with levofloxacin resistance occurred, the resumption of the growth of bacteria MIC of0.156±0.581. |
