Objectives:To investigate the clinical significance of the miRNA differential expression and miR139-5p related Notchl gene detection in colorectal cancers.Materials and Methods:Total14patients with recently diagnosed CRC, including2cases of well differentiated,10of moderately differentiated, and2of poorly differentiated. Total RNA was extracted in these specimens and converted into cDNA. Total3441of human miRNAs were detected in CRC tissues by microarray. According to the microarray results, some miRNAs were selected for validating by Real-Time RT-PCR. The target genes were searched through the TARGET SCAN website. Additionally, Notchl gene protein expression level was detected in normal colon tissues and CRC by using EnVision method of immunohistochemistry, and the clinicopathological significance of Notchl protein overexpression in CRC was also analyzed.Results:194miRNAs were significantly upregulated and38miRNAs were downregulated which detected from total3441of human miRNAs in CRC tissues by microarray. The significantly upregulated miRNAs are hsa-miR-31, hsa-miR-105, hsa-miR-196b, and so on. The significantly downregulated miRNAs are hsa-miR-139-5p and hsa-miR-215. MicroRNA-139-5p associated protein Notchl is located in the cytoplasm, and the positive rate was57.3%(43/75) and71.1%(27/38) in CRC and normal tissue on immunohistochemistry, respectively (P>0.05). The strongly positive rates of Notchl were55.3%(21/38) and21.6%(8/37) in non-metastatic and metastatic CRC, respectively (P<0.05). For the TNM clinical stages, the strongly positive rate of Notch1protein was 57.9%(22/38) in early clinical stage of CRC, but only21.6%(15/56) in late stage cases (P<0.05). Additionally, the strongly positive rate of Notchl protein expression was significantly higher in≥5years disease free survival cases (59.4%,19/32) than it in<5years disease free survival cases (23.3%,10/43)(P<0.05). For WHO’s pathological differentiation, the strongly positive rate was36.0%(9/25),44.4%(12/27) and34.8%(8/23) in well, moderately and poorly differentiated CRC, respectively (P>0.05).Conclusions:The significant miRNAs differential expression profile was found in colorectal cancer compared with the patched peripheral normal tissues. Therefore, the detection of micro RNAs differentially expressed will provide a theoretical reference for colorectal cancer screening and early diagnosis. Additionally, microRNA-139-5p associated protein Notchl may become an important indicator to evaluate clinical prognoses in colorectal cancer.
