| Objective:To genotype polymorphisms of g.-964T>C in the IL-27p28gene promoter region, g.2905T>G in the second exon region,g.4730T>C in the fourth exon region and detect the association of these SNPs with asthma susceptibility in a chinese korean nationality population. Methods:The definite DNA were propagated by polymerase chain reaction(PCR). SNP Loci were monitored by single nucleotide extension method. Genotypes of these three loci were recorded. The SHEsis was used to detect deviation from Hardy-Weinberg equilibrium, to make sure whether the control group can stand for a Chinese Korean nationality population. Genotype frequency and allele frequency between the case group and the control group were compared by the table2×2.2x3of chi-square test through SPSS16.0, P<0.05indicates a statistical significance. Results:SHEsis test shows all genotype distribution of these3loci fitted Hardy-weinberg Law. T/Tã€T/C and C/C appeared in g.-964of IL-27p28gene promoter region, the P values of genotype frequency and allele frequency between the case group and the control group were0.000and0.000respectively; T/T and T/G appeared in g.2905of the second exon region, the P values of genotype frequency and allele frequency between the case group and the control group were0.451and0.418respectively. T/Tã€T/C and C/C appeared in g.4730of the fourth exon region,the P values of genotype frequency and allele frequency between the case group and the control group were0.097and0.069respectively. Conclusions:IL-27p28gene polymorphism of-964T>C might be associated with asthma susceptibility in a Chinese Korean nationality population, and the polymorphisms of g.2905T>C and g.4730T>G might be no association with asthma susceptibility in a Chinese Korean nationality population,but it does not rule out the association between these two loci with asthma in other population, and not rule out the possibility of other SNPs of IL-27p28might be associated with asthma susceptibility. |
