Purpose:Exercise-induced oxidative stress has been found more than30years,but The mechanisms of exercise-induced oxidative stress are not well understood.Numerous studies have confirmed that exercise can lead to the generation of superoxide anion in vivo studies have shown that has also led to enhanced skeletal muscle nitric oxide synthase activity and thus increased production of nitric oxide,nitric oxide and superoxide rapid response the formation of peroxynitrite. Peroxynitrite is a strong oxidant,we speculate it to the generation of exercise-induced fatigue plays a vital role.We create an exhausting exercise model for rats which can be induced to such fatigue.By observing related indices in blood serum and degrees of metabolism in rats, in order to more systematically and deeply, proven whether exhaustive exercise serum ONOO-induced excess generated in a system to provide a new theoretical basis for the free radical theory of movement exercise fatigue.Methods:32six-weeks-old male Kunming SD rats are divided into two groups with16rats in each group. One is the exhausting exercise group (Group E) and the other is the control group (Group C). Sampling detected immediately after exhaustive.Result:(1) After exhausting exercise, the CK activity of Group E increases14%(P<0.01). At the same time, LDH content of Group E only increases14%compared with Group C which has no significance difference.(2) After exhausting exercise, the MDA content of Group E increases42%compared with Group C,the SOD decreases300%.(P<0.01)(3) After exhausting exercise, the NO content of Group E increases8%,ONOO-increases18%dramatically(P<0.01).Conclusion1. Exhaustive exercise lead to the increase in serum CK activity, indicating that the skeletal muscle cell damage, a large number of skeletal muscle cells of CK leakage into the blood, cause of exercise-induced fatigue.2. Exhausting exercise make bodies generate excessive free radicals which can strengthen the cellular lipid peroxidation to produce MDA. Meanwhile, the decrease of SOD reflect its function to clear out the free radicals. Changes in these two indices indicate that exhausting exercise can lead to the over-production of free radicals.3. The peroxynitrite large production maybe beceause of O2·, it can combines superoxide anion NO to generate peroxynitrite which may be one of the mechanism of sports fatigue.
