Object We studied the protein and mRNA level of the disrupted-in-schizophrenia1(DISCI) and the phosphodiesterase4(PDE4) in the hippocampus of epileptic mice induced by pilocarpine to explore its interaction in the pathogenesis of epilepsy.Methods1.246Male C57BL/6mice were randomly assigned into experimental group (n=198) and control group(n=48).The mice in experimental group were given small dosage of pilocarpine to set up the epileptic mice model.2.Expression of DISCI, PDE4, phosphodiesterase4B (PDE4B) and minichromosome maintenance protein2(MCM2) in the hippocampus of epileptic mice was analysed via immunohistochemistry and real-time PCR at different time points (3,7,14,28days) after status epilepticus.Results1.DISC1protein mainly located within the hippocamal formation nucleus and cytoplasm.The expression level of DISCI protein and mRNA level had no significant difference in control group at each time points (P>0.05).Compared with the control group, the expression level of DISCI in experimental group had no significant difference at the time point(3days) but was significantly lower from7days to21days, which presented a deceasing tendency.2. PDE4and PDE4B protein mainly located within the hippocamal formation cytomembrane and cytoplasm.The expression level of PDE4B protein、mRNA and PDE4protein had no significant difference in control group at each time points (P>0.05).Compared with the control group, the expression level of PDE4in experimental group was no significant difference at the time points of3days and7days but was significantly lower at the time point of14days, and was significantly higher at the time point of28days, which appeared to have an ascend trend after the initial decline. A significant correlation existed between the expression level of DISCI and PDE4in the early time(<14days)(P<0.01)3.We found that the quantity of MCM2+cell and MCM2mRNA in control group was significant different at each time point, which appeared a decreasing tendency.Compared with the control group, the expression level of MCM2in experimental group was significantly higher at each time point, which appeared to have an ascend trend in the proliferation rate. A significant correlation existed between the expression level of DISC1and MCM2(P<0.01)Conclusions1.DISCI might participate in the pathogenesis of epilepsy by mediating the proliferation of progenitor cells.2.PDE4and PDE4B might get involved in the pathogenesis of epilepsy by mediating the proliferation of progenitor cells through DISCI in the early phase.
