| Objective:To investigate the effect of lipopolysaccharide and IKKβ inhibitors on expression of IKKβ,IκBα, P53and P21protein in cervical cancer cell lines, Siha and C33A.Methods:Use LPS and SC-514to stimulate on the cervical cancer cell lines Siha and C33A, and then the method of Western Blot was used to detect the expression of IKKβ,IκBα, P53and P21protein in cells.Results:1ã€After the stimulation of lipopolysaccharide on Siha, the changes on the expression of the IKKβ,IκBα, P53and P21protein.In the HPV16-positive Siha cells, following the treatment of lipopolysaccharide in35min, the relative expression levels of IKKβ protein were increased, before and after the stimulation were:0.5469±0.05682,1.2774±0.08809,respectively,and the differences existed between them were statistically significant (P<0.05); And the relative expression levels of IκBα protein were decreased,were0.44437±0.25656ã€0.08400±0.04850respectively,and the differences existed between them were statistically significant (P<0.05); And the relative expression levels of P53protein were decreased,were0.2244±0.03631ã€0.0699±0.01215, respectively,and the differences existed between them were statistically significant (P<0.05); And the relative expression levels of P21protein were decreased,were0.2610±0.02577ã€0.0657±0.01643, respectively,and the differences existed between them were statistically significant (P<0.05). 2ã€After the stimulation of lipopolysaccharide on C33A, the changes on the expression of the IKKβ,IκBαã€P53and P21protein.The HPV16/18negative C33A cells,following the treatment of lipopolysaccharide in35min, there were no apparent changes on the relative expression levels of IKKβ protein,before and after the stimulation were:0.8003±0.29221,1.0759±0.24533,respectively,and the differences existed between them were not statistically significant (P>0.05); and there were no apparent changes on the relative expression levels of IκBα protein,before and after the stimulation were:0.6239±0.13702ã€0.7153±0.08665,respectively,and the differences existed between them were not statistically significant (P>0.05);and there were no apparent changes on the relative expression levels of P53protein,before and after the stimulation were:2.5848±0.42433ã€2.5705±0.45800,respectively,and the differences existed between them were not statistically significant (P>0.05);and there was no expression of P21on C33A cells.3. After the stimulation of SC-514on Siha, the relative expression of the IKKβã€IκBαã€P53and P21protein.In HPV16-positive Siha cells,Firstly stimulated on the cells with lipopolysaccharide in35min, and then treated them with SC-514respectively in0ã€60min, the relative expression levels of IKKβ protein were decreased, before and after the stimulation of SC-514were:0.2172±0.04407ã€0.0789±0.02192,respectively,and the differences existed between them were statistically significant (P<0.05); And the relative expression levels of IκBαprotein were increased,were0.1343±0.02672ã€0.2663±0.02261,respectively,and the differences existed between them were statistically significant (P<0.05); And the relative expression levels of P53protein were increased,were0.1159±0.00711ã€0.3343±0.08890, respectively,and the differences existed between them were statistically significant (P<0.05); And the relative expression levels of P21protein were increased,were0.0258±0.00826ã€0.1865±0.03124, respectively, and the differences existed between them were statistically significant (P<0.05).4. After the stimulation of SC-514on C33A, the relative expression of the P53and P21protein.The HPV16/18negative C33A cells, Firstly stimulated on the cells with lipopolysaccharide in35min, and then treated them with SC-514respectively in0ã€60min,there were no apparent changes on the relative expression levels of P53protein,before and after the stimulation of SC-514were:2.1650±0.14691ã€2.1396±0.21367, respectively,and the differences existed between them were not statistically significant (P>0.05); and there was no expression of P21on C33A cells.Conclusion:1. Lipopolysaccharide can increase the expression of IKKβ protein and decrease the expression of IκBα protein expression in HPV16-positive Siha cells, but cannot affect the expression of IKKβ and IκBα protein HPV16/18negative cells;2. IKKβ inhibitor can reverse the effect of the lipopolysaccharide on HPV16-positive cervical cancer cells about the expression of IKKβ and IκBα protein;3. Following the stimulation of lipopolysaccharide on the HPV16-positive cervical cancer cells, the expression of P53and P21protein decline obviously; But there is no obvious effect on C33A;4. IKKβ inhibitor can reverse the effect of the lipopolysaccharide on HPV16-positive cervical cancer cells about the expression of IKKβ and IκBα protein; But there is no obvious effect on C33A. |
