Objectives:To explore the protective effects of bFGF on motor endplate degeneration and spinal cord anterior horn motoneurons after experimental contusive spinal cord injury (SCI) at rats, and analyze the possible mechanism of its protective role.Methods:Fourty-five rats, weighted200-250g, were randomly assigned to three groups:group A (sham operation group, n=5), group B (experimental group, n=20), each rat received injection of bFGF, and group C (the control group, n=20), each rat received injection of NS. The rats of B、C group were assigned to B1、B2、B3、B4group and C1、 C2、C3、C4group at1、2、4、8week after SCI, respectively. All the rats of B、C group were received injection from subarachnoid guide drug possession after SCI. and the rats of A group were treated with the lamina open alone. BBB score was enforced on each group, the content of Spinal cord tissue and the anterior tibial muscle were detected with HE staining and the expressions of CGRP、AchE were then determined with immunohis-tochemistry.Results:1.BBB score:B2>C2, B3>C3, B4>C4(P<0.05); C4>C3>C2(P<0.05).2. HE staining:Compared with group C1, the inflammatory response of group B1was significantly milder; compared with group C2, the inflammatory response and Neuron atrophy of group B2was significantly milder; group B3were recovered better than C3, and the scar formation of group B4was significantly less than group C4.3. CGRP OD:Spinal cord tissue:B1>C1, B2>C2, B3>C3, B4>C4(P<0.05); C4>C3>C2(P<0.05); anterior tibial muscle:B2>C2, B3>C3, B4>C4(P<0.05); C4>C3>C2>C1(P<0.05).4.AchE OD:Spinal cord tissue:B3>C3, B4>C4(P<0.05); anterior tibial muscle:B3>C3, B4>C4(P<0.05); C4>C3>C2(P<0.05).Conclusion:1. bFGF can prevent the injury of spinal cord anterior horn motoneurons after SCI may be related to reduce the spinal cord remote inflammatory response. 2. Through preventing the decline of CGRP、AchE in spinal cord anterior horn motoneurons after SCI, and enhancing CGRP、AchE expression, bFGF can avoid the degeneration of motor endplate and promote their recovery.
