Objective:To investigate part mechanism of Kidney-reinforcing and Blood-activating recipe Yizhi Jiannao granule on preventing and curing Alzheimer’s disease by observing the effect of YCF on AD rats’learning and memory ability, the expression of Nrf2and8-iso-PG F2a in the hippocampus of AD rats.Methods:40SD male rats were randomly divided into four groups, the normal group(n=10), sham operation group(n=10), model group(n=10), and the Kidney-reinforcing and Blood-activating group (n=10). By injecting D-galactose into abdominal cavity combined with injecting Aβ25-35into both sides of hippocampus of the last two groups to make AD models. The same volume of Normal saline were injected in hippocampus of sham operation group in the same way, the normal group deal with nothing, All the groups were tested with Morris water maze test (MWM) after operation to test whether AD models were successful or not. Rats of Kidney-reinforcing and Blood-activating group were given with yizhi jiannao granule while the other3groups were given double-distilled water for8weeks. All the groups were tested with Morris water maze test (MWM) again when finished all the treatment above, in order to observe the change of the all rats’learning and memory ability. Finally, the hippocampus were taken out to detect the expression of Nrf2with RT-PCR, and8-iso-PG F2α with ELISA.Results:1. Behavioral results: In Morris water maze test, the model group、the Kidney-reinforcing and Blood-activating group had longer escape latency, and also can shorten the time staying in the original platform as well as decreased the average frequency of passing through the hidden platform compared with the normal group. After the intervention of Kidney-reinforcing and Blood-activating therapy, the Kidney-reinforcing and Blood-activating group can significantly shorten the escape latency, increase the stay time in the former platform quadrant and former platform crossing times compared with model group. It explained that the Kidney-reinforcing and Blood-activating therapy can improve the learning and memory ability of AD rats effectively.2. RT-PCR results:Compared with normal group, the expression of Nrf2decreased obviously in the model group (P<0.05), however, there is no significant difference between sham operation group and normal group (P>0.05). Compared with model group, the expression of Nrf2increased significantly in the Kidney-reinforcing and Blood-activating group (P<0.05).3. ELISA results:Compared with normal group, the expression of8-iso-PG F2α increased obviously in the model group (P<0.05), but there is no distinct difference between sham operation group and normal group (P>0.05). Compared with model group, the expression of8-iso-PG F2α decreased evidently in the Kidney-reinforcing and Blood-activating group (P<0.05).Conclusion:1. By injecting D-galactose into abdominal cavity combined with injecting Aβ25-35into both sides of hippocampus of the last two groups can make representative AD rats model.2. The Kidney-reinforcing and Blood-activating recipe Yizhi Jiannao granul can effectively improve the learning and memory ability in the AD rats model.3.The change of expression of Nrf2and8-iso-PG F2α in the AD rats model may be one of the pathogenic mechanisms of Alzheimer’s disease.4. Yizhi Jiannao granule maybe treat AD by increasing the expression of Nrf2and decreasing the8-iso-PG F2α, thus it can reduce the oxidative stress.
