Objective:To elucidate the mechanism of the separated groups from Bizhongxiao decoction(BZXD) in treatment of rheumatoid arthritis(RA) by analyzing the effects of the separated group1(Antipyretic Detoxicate group)、separated group2(Dissolving Dampness group)and separated group3(Activating Blood Circulation group) from BZXD on ERK1/2and ERK5expressions in synovium of CIA rats.Methods:The experiment arthritis rat model was established by subcutaneous injection of II collagen and complete Freunds adjuvant. ERK1/2and ERK5expressions at different times in five groups were detected by Western blotting,and semi-quantification analysis was performed with gray scale scanning software.Results:ERK1/2、ERK5expressions of synovium in model group on the21th day were higher than those in normal group(P<0.05). ERK1/2、ERK5expressions of synovium in model group on the28th day were higher than those on the21th day(P<0.05). After separated group1、separated group2and separated group3treatments,ERK1/2、 ERK5expressions were sigificantly lower than those in model group at the different timepoints(P<0.05) and decreased gradually(P<0.05). ERK1/2expression of synovium in the separated group2decreased more significantly than that in the separated group1and separated group3(P<0.05), ERK5expression of synovium in the separated group1decreased more significantly than that in the separated group2and separated group3(P<0.05).Conclusion:During the process of CIA, with the increasing severity of arthritis, ERK1/2and ERK5edged up. It is suggested that ERK1/2、ERK5are related to CIA synovitis and RA. The separated groups from BZXD all can edge down ERK1/2、ERK5expressions in synovial tissue of CIA rat, then inhibit the activation of ERK1/2、ERK5protein kinase signal transduction pathways. The separated group2can inhibit the activation of ERK1/2protein kinase signal transduction pathway more significantly than other groups, the separated group1can inhibit the activation of ERK5protein kinase signal transduction pathway more obviously than other groups, which may leads to inhibit the abnormal proliferation of synovial and articular bone erosion.
