Objective:To investigate the relationship between DNA ploidy, clinicopathological features and molecular surgical margin in oral squamous cell carcinoma (OSCC).Methods:Fresh tissues, clinicopathological data of30cases from OSCC and fresh tissues of5cases from normal oral mucosa were collected in the Third Xiangyang Hospital of Central South University and Hunan Province Tumor Hospital. DNA index (DI), DNA ploidy, S phase fraction (SPF), proliferative index (PI) of all the above mentioned tissues were detected. At the same time, the expression of ki67in OSCC lesion, surgical margin, normal oral mucosa was also detected by using flow cytometry.Results:(1) The proportion of DNA heteroploid of30cases in OSCC was63.3%(19cases), while for all the normal oral mucosa, only DNA diploid was found;(2) The expression of DI、 SPF. PI in OSCC differed significantly from that of normal oral mucosa (P<0.01);(3)There was significant difference when comparing the expression of SPF、 PI in the heteroploid group with that in the DNA diploid group(P<0.01);(4)There was a difference with statistical significance between the DNA heteroploid rate in well differentiated group and that in poor differentiated group(P<0.01); the DNA heterploid rate in clinical stage I differed from stage III, when considering clinical stage II, Ⅲ, IV as a whole, the DNA heteroploid rate in stage I differed from the result of stage II, III, IV(P<0.01). The DNA heteroploid rate in group with Cervical node metastased was not similar to that of group without cervical node metastased;(5) The expression of ki67in surgical marginal tissues in DNA diploid group of OSCC was no clear interrelationship with result from DNA heteroploid group (P>0.05). The expression of ki67decreased gradually according to the increase in distance of tissues between tumor and surgical margin.Conclusions:Based on the conducted research, we could conclude that:(1) The type of DNA ploidy in OSCC is related to histological differentiation of tumor, clinical staging, Cervical node metastased;(2) There is no correlation between the type of DNA ploidy and ki-67expression in surgical margin;(3) According to the different expressions of Ki67in different margins,1.5cm distant from the tumor’s margin could be considered as molecular negative margin.
