| Objective:In recent years, research has found that the electrical coupling mediated synchronization mechanisms play an important part in epileptic electricity activities, and gap junctions (GJ) acted a very important role in this process.The experiment observed the changes of behavior and EEG in seizure rats induced by4-AP, investigations of the expression of Cx36and Cx43in hippocampus by the methods of HE staining, immunohistochemical staining, Western blot, to further understanding of the role of Cx36and Cx43in the mechanisms of epileptogenesis.Methods:Using brain stereotactic apparatus to implant electrodes, the seizure model was prepared on intraperitoneal injection with4-aminopyridine. forty-eight SD rats were randomly divided into4groups. There were blank control group,4-AP induced seizure model of the8st hour,24rd hour, and72th hour.Investigating of the expression of Cx36and Cx43in hippocampus.Results:1.Cx36is expressed in cortex and hippocampus(mainly in the neurons), CA1, CA3and dentate gyrus. There was a significant difference than that in control group at the same time point(P<0.05).The expressions of Cx36in hippocampus neurons increased firstly, and reached peak at24hour, after gradually declined in4-AP induced seizure model group. 2. Cx43is expressed in cortex, hippocampus and brain stem (mainly in astrocyte), CA1and CA3. There was a significant difference than that in control group at the same time point(P<0.05).The expressions of Cx43in astrocyte increased gradually in4-AP induced seizure model group.Conclusions:1.The expression of Cx36in hippocampal neurons in seizure rats induced by4-AP was significantly increased, indicating that may participate in the pathogenesis of epilepsy.2.The expression of Cx43in astrocyte in seizure rats induced by4-AP was differently, indicating that may participate in the reconstruction of the function of electrical synapse. |
