Identification PSME1as The Interactive Protein Of LOH12CR1

Objective:Our reaserch group has found that LOH12CR1(Loss of heterozygosity12chromosomal region1protein) is the interactive protein of C-ternimal regional protein of LKB1/STK11in2008.LOH12CR1,identified on chromosome12,is extremely conservitive in evolution, LOH12CR1Subcellular Location at endochylema and nucleus. The function of LOH12CR1is unknown. Our reaserch group has screened the PSME1(Proteasome activator complex subunit1)as the interactive proteins of LOH12CR1.PSME1,which encoded prosome activator subunit1(PA28a),mainly Enhance the protein-degragation function of proteasome and participated in MHC-I restricted antigen presention. Our reaserch will prove the relationship between LOH12CR1and PSMEl,and provide new orientation for the study of LOH12CR1.Method:With instrant contransfect HEK293T cell with pCDNA3.1-myc-LOH12CR1and pCMV-tag2B-PSME1, co-immunoprecipitation assay and immunofluorescence,we aim to identify the interaction between LOH12CR1and PSME1and co-immunolocaliztion of LOH12CR1and PSME1.Result:1.We observed that the PSME1can interactive with LOH12CR1.2. We observed that the PSME1can co-locate with LOH12CR1Conclution:As the interactive protein of PSME1, LOH12CR1can be probably participated in the protein-degragation function of proteasome and MHC-I restricted antigen presention. This hypothesis need to be proved in our further reseach.
