ObjectiveExplore the ApoE gene polymorphism and sporadic Alzheimer’s disease(AD) cognitive function relationship.MethodSelect in August2010to January2011in Hunan Province sporadic Alzheimer’s disease a total of100cases over the same period from the hospitalized non-Alzheimer’s disease healthy elderly subjects a total of102cases for study. Comparative study, all subjects underwent the Mini-Mental Status Examination (MMSE), Hachinski Ischemic Index Scale (HIS) score, the score of the Wechsler Memory Scale (WMS) and the Wisconsin Card Sorting Test (WCST). Polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) Determination of the ApoE gene polymorphism on the subjects.Adopt SPSS13.0for statistical analysis.Results1. AD group and normal group of patients in age, gender was no significant difference (P>0.05); patients of two groups in terms of history of hypertension, diabetes mellitus, coronary heart disease history was no significant difference (P>0.05); smoking history, drinking history, the two groups of patients were significant differences (P>0.05); serum lipid levels in AD patients, diastolic blood pressure compared with the control group, the difference was statistically significant (P<0.05).2. There was no significant difference between AD group and control group in AST (glutamic-oxal(o)acetic transaminase), ALT(glutamate-pyruvate transaminase), BUN(urea nitrogen) UA(uric acid),TG (triglyceride), HDL-C (high density lipoprotein-cholesterol), LDL-C(low-density lipoprotein-cholesterol) ApoA-I, ApoB (apolipoprotein B)(P>0.05); However, there was significant difference between AD group and control group in cholesterol levels (P<0.05).3. Compared with the control group:the score of the AD group was significantly lower in cognitive function of Wechsler Memory Scale, Wisconsin Card Sorting Test (P<0.05); the classification number, the correct number of the AD group was decreased significantly in Wisconsin Card Sorting Test (WCST)(P<0.05); the continuous reaction, continuity error, non-perseverative errors of the AD group was significantly increased (P<0.05).4. Compared with the control group:the distribution of frequency of ApoEε3/4genotype in the AD group is greater(P<0.05). the distribution of frequency of ApoEε3/3genotype in the AD group is less(P<0.05). ApoEε3allele frequencies in the AD group was lower (P<0.05), ApoEε4allele frequency in the AD group was higher (P<0.05).5. Compared with the control group:the level of serum TC in the AD group was higher; what is more, the level of serum TC of ApoE s4carriers was higher than those without ApoEε4(P<0.05).6. In accordance with the long-term memory, short-term memory and transient memory of the three memory types in Wechsler Memory Scale, there was no difference in long memory between the ApoE ε4allele(P>0.05), but significant different (P<0.05) in short-term memory, transient memory compared with the other allele(ε2, ε3); AD patients carrying different alleles in the Wisconsin card sorting test of the indicators were not significant different (P>0.05), and the sustained response of individual who carried ε4allele is greater than the other gene carriers, but there was no significant difference (P>0.05).7. The logistic regression analysis with the limitation of age and sex showed that the ApoEs4allele is the independent risk factor of sporadic AD(OR=2.69,P=0.002,95%CI=1.78-5.26)Conclusion1. There was a relationship between sporadic Alzheimer’s disease and ApoEs4. What is more, ApoEε4was a single risk factor for AD.2. Apoe s4allele can influence individual short-term memory and short-term memory, but there no influence to long-term memory.
