[Abstract] Objective: To confirm the problems of learning and memory are relevant tothe effect on the expression of N-methyl-D-aspartic acid (NMDA)receptor in offspringSD mice hippocampus which caused by infection with HCMV during early pregnancy。Methods:Eight-week-old female SPF SD mice (n=18)were matched with malerats(3:1). Pregnant rats were randomly divided into control group and experimentalgroup, each group contained9rats。 Pregnant rats in experimental group wereintraperitoneal injected of0.8mL5x107L-1TCID50(50percent tissue culture infectivedose) HCMV suspension, and respectively, pregnant rats in control group wereintraperitoneal injected the same amount of human embryonic fibroblast supernatant on5gestation days. Obtained the brain of rat offspring(n=10)30days after birth randomly,then observed the morphology of hippocampus by hematoxylin eosin staining;Theability of learning and memory was tested by Morris water;The expression of NMDAreceptor in hippocampus was determined by immunohistochemical staining。Result:The number of hippocampus nerve cells in control group did not reduce,cells arrangedcompact and well-distributed,nucleus was round or oval, nucleolus was obvious,chromatin was even, compared with control group,the pathological changes such asdegeneration、necrosis、edema and nucleus disappearance of nerve cells and giant cellswere found in infected group under light microscope。In Morris water test,learning andmemory dysfunction was observed in infected group(P<0.05);In immunohistochemicalstaining,relative absorbance of NR1、NR2A、NR2B receptor decreased significantly ininfected group(P<0.05)。Conclusion: The infection with HCMV during earlypregnancy can result in damages of learning and memory in offspring mice,whichmight be relevant to its regulating effect on the expression of NMDA receptor inhippocampus。...
