Objective T helper cells of Thl, Th2, Th17, CD4+CD25+Foxp3+regulatory T cells (Treg) and lymphocyte subsets in peripheral blood were detected by flow cytometry in children with asthma and pneumonia. Analyse the similarities and differences of the detected cytokine,more importantly, in order to explore the significance of CD4+T cells and lymphocyte subsets in children with asthma and pneumonia.Methods Questionnaires were made according to the literature and previous studies, questionnaire surveys the children with asthma or pneumonia in the pediatric ward, during September2010to February2012.We adopt the strict enterer and elimination standard, screening fifteen patients with asthma and seventeen children with pneumonia were enrolled. In addition, fifteen health children were enrolled as control group at the same time. The fasting venous blood samples2ml were taken on the morning, and cope with the samples within4hours. By flow cytometry detected the cytokine of Thl, Th2, Th17, CD4+CD25+Foxp3+regulatory T cells and lymphocyte subsets. The clinical relative factors and experimental data were statistically analyzed using SPSS18.0statistical software, each set of experimental data expressed as mean±standard deviation, in each experimental group were analyzed using single factor analysis of variance, multiple compared with LSD method; correlation analysis using Spearman method, P<0.05was considered statistically significant. Results Result1The children with asthma group peripheral blood CD4+CD25+Foxp3+regulatory T cells(7.58±2.75)%in CD4+T were higher than the pneumonia group (3.76±1.47)%and control group (5.88±1.28)%, there was remarkably statistical difference(F=14.041, P=0.000). There was no remarkably statistical difference of Thl in three groups(P>0.05).The percentages of Th2(4.34±2.08)%of asthmatic children were remarkably higher than those in the control(2.01±0.91)%and pneumonia group[(1.61±0.74)%](F=6.685, P=0.003), Th17of asthmatic children(1.64±1.01)%were remarkably higher than those in the control (0.95±0.45)%and pneumonia group[(0.94±0.51)%](F=4.590,P=0.016). The ratio of Th1/Th2(1.83±0.77)%of asthmatic children were remarkably lower than those in pneumonia group[(3.38±1.39)%](F=5.202, P=0.010),but Thl7/Treg (0.36±0.23)%of pneumonia group were remarkably higher than those in control group(0.19±0.11)%(F=4.052, P=0.022).Result2The levels of CD3+of asthmatic children were lower than those in the control group(F=2.772,P=0.040);while there is a remarkably higher percentages of CD4+in children with pneumonia or asthma than those in the control group(F=3.301,P=0.031,0.037). The levels of CD16+56+of asthmatic children were lower than those in the control and pneumonia groups(F=3.833,P=0.041,0.011),but the levels of CD19+and CD4+/CD8+of asthmatic children were higher than those in the control and pneumonia groups(P<0.05),there was remarkably statistical difference.There was no remarkably statistical difference of CD19+and CD4+/CD8+between pneumonia and control groups.Result3The proportion of neutrophils and leukocyte was no significant difference among the3groups (P>0.05).The percentages of lymphocytes in asthmatic group were remarkably lower than the control group(F=5.022,P=0.003); The percentages of monocyte cells in asthmatic group were remarkably higher than the control group, while the percentages of eosinophilic cells in asthmatic group(6.22±3.20)%were remarkably higher than the control(2.62±0.79)%and pneumonia group [(0.47±0.29)%](P<0.01).Result4There was a remarkably correlation between blood routine,CD4+T cells and Lymphocyte Subsets.Conclusion T helper cells (Thl/Th2/Thl7) in CD4+T cells and Lymphocyte subsets by flow cytometry could be used as indicators of immune reaction in children with asthma or pneumonia.The result of this study indicated that there is the unbalance of immune in children with asthma or pneumonia,but there was remarkably difference between the two groups. Th2, Thl7, CD4+CD25+Foxp3+regulatory T cells,CD19+,CD4+/CD8+,percentages of monocyte and eosinophilic cells in asthma group were all higher than the control group; and Th1/Th2,CD3+,CD4+,CD8+,CD16+56+in asthma group were all lower than the control group. But compared with normal group there were only CD4+CD25+Foxp3+Treg,Thl7/Treg,CD4+,percentages of monocyte and eosinophilic cells in pneumonia group having the change. Above all,we can see the clinical significance of the detected cytokine in our study about diagnosis,treament and observation of clinical effect in the two diseases.
