| Objective:To observe the effect of ethyl acetate extract of sappanwood on the content of transforming growth factor betal in blood serum of rats with chronic rejection after allogenic cardiac transplantation and observe the pathomorphology of transplanted heart.To preliminarily discuss the mechanis m of the ethyl acetate extract of sappan wood resist the chronic rejection.Methods:The allogenic cardiac transplantation model was established by using the improved Ono operation kill and with the male Wistar rat as donor and the male SD rat as receptor. The successful opration rats were given CsA solution35mg/(kg-d) from the method of intragastric administration for21days.The rats were divided into three groups randomly after21days.CsA group was given CsA solution12mg/(kg·d). Sappanwood group was given the ethyl acetate extract of sappanwood solution736mg/(kg·d).Model control group was given olive oil depending on the weight of rats. The blood sample and transplanted heart were took after7days’dosing.The content of TGF-β1in blood serum was tested by ELISA, and the pathomorphology of transplanted heart was observed by light microscopic.Results:The content of TGF-β1in blood serum of rats was reduced obviously in sappanwood group and CsA group. The two groups compared with model control group with significant difference(p<0.01). The CsA group content lower than sappanwood group and the difference was not statistically significant (p>0.05).Compared with model control group, the pathological damage of the transplanted heart was reduced in sappanwood group and CsA group.Conclusion:1.The ethyl acetate extract of sappanwood can reduce the content of TGF-β1in blood serum of rats of chronic rejection after allogenic cardiac transplantation.2. The ethyl acetate extract of sappanwood and reduce the myocardial pathological damage of rats of chronic rejection after allogenic cardiac transplantation. |
