Objective The objective of this work is to investigate the expressions of HA117in non small cell lung cancer and the relationship of their expressions and the pathological stage of NSCLC.Materials and MethodsSamples28specimens of primary non small cell lung cancer were obtained from patients who had surgery in the Qilu hospital of Shandong University between2008and2010, fixed in10%paraformaldehyde, embedded in paraffin.ReagentThe Ambion RecoverAll Total Nucleic Acid Isolation kit was bought from OIAGEN company. The M-MLV reverse transcriptase enzyme was bought from PROMEGA company. The2×SYBR Premix DimerEvaser, Dnase I (Rnase Free)、 PCR primer、DL2000DNA Marker were bought from TAKARA company。MethodThe expressionsof HA117gene in the28NSCLC specimens and7corresponding normal lung tissue specimens was performed by qRT-PCR.Statistical analysisAll data are processed by SPSS17.Ostatistical software, quantitative data of two groups was used to compare with the X2test. Survival analysis are performed by Kaplan-Meier method. Significance level is P<0.05.ResultThe HA117mRNA relative expression quantity is higher than that in cancerous tissues。There were significant differences of HA117mRNA expression between NSCLC tissues and lung control tissues(P=0.042). The HA117mRNA relative expression levels were significantly higher in older people than in young (P=0.037), higher in nonsmoker than in smoker (P=0.025), higher in adenocarcinoma than in squamous carcinoma (P=0.022). However, the HA117mRNA levels were not significantly different in patients with gender or whether the tumor had lymph node metastasis status. No significant difference was seen between any pathologic stages of lung cancer. There was no difference of cancer recurrence (P=0.372) and prognosis (P=0.605) between the group of low HA117mRNA levels and the group of high HA117mRNA levels (Log-rank test)。ConclusionExpression of HA117mRNA was significantly high, in comparison with normal lung tissue.These data suggest that in older people(>60), smoked with adenocarcinoma, increased expression of HA117might have an important role in NSCLC.There was no significant relevance between the expression level of HA117mRNA and cancer recurrence or the prognosis.High levels of HA117related protein expression in lung neoplasm tissue indicate a strong resistance to ATRA.
