OBJECTIVEFruit of Chaenomeles is commonly used as Chinese traditional medicine. On the market, four kinds of fruit of Chaenomeles are used as traditional medicine. The only fruit of Chaenomeles in Ch.P is called Chaenomeles species (Sweet) Nakai. It is mainly produced in An Hue, Zhe Kiang, He Nan, Hun Bee, Si Chuan and Shan Dong province in our country. Lin yi and He ze areas in Shandong province, are the two largest planting areas of Chaenomeles, Fruit of Chaenomeles of Lin yi has not been taken more attention because of lack of the researches of quality controls. In order to make better use of the resources of the Chaenomeles planted in Shandong province, we used modern chromatographic method such as HPLC-DAD-MS to evaluate the qualities of the fruits of Chaenomeles as Chinese traditional medicine.METHODCollecting many batches of fruits of Chaenomeles from three different provinces producted in2011and2012.A reversed-phase HPLC method was developed for determination of oleanolic acid and ursolic acid in fruits of Chaenomeles.Extracting with water and using reversed-phase HPLC method to establish the fingerprints of the fruit of Chaenomeles. Combining with principal component analysis, cluster analysis and similarity analysis to estimate the fingerprints. The method has high precision and repeatability.Using HPLC-DAD-MS technology to study and speculate the possible structures of common peaks in the fingerprint.Using HILIC method to research the water-soluble components of the fruit of Chaenomeles for the first time.RESULTSThe determinations of content of OA and UA in fruit of Chaenomeles show that all the samples collected are quality qualified. Accorroding to compare of the content of OA and UA in fruit of Chaenomeles produced in2011and2012, we know that the content of OA and UA in fruit of Chaenomeles could reduce by years. In addition, processing methods may affect the content of the OA and UA in fruit of Chaenomeles, so the processing method should be further standardized especially to Shandong Chaenomeles.HPLC fingerprint used for the quality control of fruit of Chaenomeles has been established. Principal component analysis, cluster analysis and similarity analysis results show that fruit of Chaenomeles from different provinces have similar quality.Using HPLC-MS detecting technology, we got possible structures of six compositions in the fruit of Chaenomeles. HILIC method was also developed.CONCLUSIONThe topic selected the water-extracted part of fruit of Chaenomeles which has modern pharmacological activities as the research object. The research work adopts new modern chromatographic analysis technologies to establish the fingerprints. Combination with determination of the content of OA and UA, the comprehensive quality control method for fruit of Chaenomeles has been established and been used to evaluate the quality of Shandong fruit of Chaenomeles. The research results will be helpful to promote the development of Shandong fruit of Chaenomeles as medicinal resource.
