Objective:Study the influence of Yunpiheluo Granule_on the proliferation of rat schwann cells cultured in high glucose and expression of NGF, as well as the spleen and collateral effect of particles on the indicators, in order to clear the mechanism of prevention and treatment of diabetic peripheral neuropathy, provided for the clinical application of experimental basis.Methods:Experiments using rat Schwann cells in culture and in vitro serum pharmacological method, a blank unit, model groups, the spleen, and complex particle Group (low, medium and high doses), mecobalamin Group (control group of WM), Tang Mai Kang Group (control group of traditional Chinese medicine). Drugs on rat Schwann cells cultured in high glucose, tetrazolium salt (MTT) test24h,48h, and72h of different groups at different time of Schwann cell OD Value; Retro virus polym erase chain reaction technology (RT-PCR) testing the drugs effects on rat Schwann cells NGF-mRNA Expression; rat Schwann cells by immunocytochemical method using on NGF Protein expression; enzyme linked immunosorbent assay (ELISA) test method of cultivation of clear liquid secretion of NGF.Results:MTT Experimental results show that high glucose inhibits proliferation of rat Schwann cells (compared with the blank group, p<0.05), Yunpiheluo Granul can counteract the proliferation inhibition effect of high glucose and there are different in time-effect and dose-effect. RT-PCR results showed that each treatment group could raise NGF-mRNA expression which was inhibits by high glucose. Results show by immunocytochemical method, spleen and complex particle can significantly improve Schwann cells under high glucose, which is significant different from model group (p <0.05). ELISA test results tell that high sugar can inhibit rats schwann cells to secrete NGF. Schwann cells NGF expression of each treatment group has increased (p<0.05). Conclusions:Yunpiheluo Granulcan restrain the inverse of the cell proliferation results under high sugar environment, promote Schwann cell NGF Gene and protein to expression, and increase NGF Secretion in cell culture supernatant, which medium doses are most obvious.
