ObjectiveTo explore the effect of nimesulide on the cell proliferation and gene expressionof P53, Sh3kbp1in TSC-6cell line.MethodsExperimen were divided into control group,200μmol/L,400μmol/Lnimesulide treatment group.Inhibition of HSC-T6cell growth induced by nimesulideat differrent concent rations was assayed by MTT method. Cell apoptosis weredetected by flow cytometry, moreover. Alterations of cyclooxygenase-2(COX-2),Sh3kbp1,P53mRNA expression were evaluated by reverset ranscription polymerasechain reaction (RT-PCR).ResultsMTT results showed the proliferation of HSC-T6cell were inhibited by200μmol,400μmol/L nimesulide treatment48h,72h after. Flow cytometry showedthat the apoptotic rate in control,200μmol/L and400μmol/L nimesulide treatedcells were respectively10.23±1.34%,25.23±7.7%,65.87±2.89%, the apoptoticrate significantly different (p <0.05). After48hours,72hours, the expression ofCOX-2mRNA in HSC-T6cells was decreased, P53and Sh3kbp1mRNA wasincreased by nimesulide (400μmol/) treatment (p <0.05).ConclusionsNimesulide can inhibit the growth of HSC-6cell line in vitro. The mechanism isprobably associated with inhibition of cell proliferation, down-regulation of theexpression of COX-2,up-regulation of the expression of P53、Sh3kbp1.
