Objectives To investigate the most suitable concentration and mechanism of theselective antitumor effect of Resveratrol and4,4’-Dihydroxybiphenyl.Methods Lung cancer cells, nasopharyngeal carcinoma cells and Human bron-chial epithelial cell were treated with0~60μM resveratrol and4,4’-dihydroxybiphenylrespectively. To find out the drug concentration range which can inhibit the growth oftumor cells but have little adverse effect on normal cells, CCK8assay was used todetect the activity of tumor cells after treated with different concentration of RES andDID respectively; The clone formation rate of different tumor cells were observedafter treated with different concentration of RES and DID;The migrating capabilityof the tumor cells were detected by transwell test after treated with dfferent concen-tration of RES and DID; The Phase distribution of cell cycle of tumor cells andnormal cells were analyzed by flow cytometry (FCM) after treated with dfferentconcentration of RES and DID respectively; apoptosis of tumor cells was measuredby DNA Ladder assay after treated with different concentration and time ofResveratrol and4,4’-Dihydroxybiphenyl; The proteins expression of CAV-1、NQO1、HSP90、ATP and PCNA were determined by Western Blot to analyze where theycan be used as the evaluation index of the selective anti-tumor effect of RES and DID.Results CCK8assay showed that7.5~30μM Resveratrol and4,4’-Dihydroxy-biphenyl could significantly inhibit the growth of different tumor cells (P<0.05), buthave little effect on the growth of16HBE. Plate colony formation assay and Trans-well test further confirmed10~30μM Resveratrol and4,4’-Dihydroxybiphenyl couldsignificantly inhibit the clone formation rate and migrating of different tumor cells. The flow cytometry analysis showed that tumor cell’s cell cycle were arrested at Sphase after treated with10~30μM RES and DID; but there is no significant change ofcell cycle of normal cells. DNA ladder test showed no DNA ladder bands after CNE-2were treated with50μM Resveratrol and4,4’-Dihydroxybiphenyl, while increasedapoptosis after treated with greater dose of Resveratrol and4,4’-Dihydroxybiphenylfor longer time (﹥24h). Western-blot analysis showed that10~30μM Resveratrol and4,4’-Dihydroxybiphenyl decreased the expression of PCNA, HSP90, ATP, andincreased the expression of CAV-1and NQO1with concentration-dependent.Conclusion1.30μM RES and DID induced cell cycle of tumor cells arresting in S phase andinhibit the growth and migrating of them, but had no influence on16HBE cells.2. CAV-1、NQO1、HSP90、ATP、PCNA can used as the evaluation index ofthe selective antitumor effect of RES and DID.
