Objective:To observe the influence of Yunnan Baiyao and total Saponins of PanaxNotoginseng using locally for the healing of anastomotic stoma in vivo, and to explorethe bacteriostatic action of Yunnanbaiyao water solublesin vitro.To prove the YunnanBaiyao andPNS can promote the healing of intestinal anastomosispreliminarily.Method:Part I: To observe the effect of Yunnan baiyao and PNS for the healing ofintestinal anastomosis with the animal experimental.45SD rats weretaken thesurgeries of bowel resection and anastomosis withthe measure ofSingle-layer interrupted Varus SutureIn process of intestinal anastomosis.Checkingwhether therewere any stomal leak before separating them into Control group, PNSGroup, Yunnan baiyao grouprandomly. Spraying saline solution on the anastomoticstoma for the control group,YunanBaiyao for Yunnan baiyao group, and the PNS forthe PNS group. To estimate the effects of two drugs on the anastomosis withcomparing the Burst pressure of their anastomosis, hydroxyproline content andanastomotic HE staining, Masson staining, the VEGF/CD31immunohistochemicaland histological changes.Part II: To study the antibacterial activity of the water soluble substances ofYunnan Baiyao.6mm diameter of round sterilization paper were soaked overnight inseveral different concentration of water solution of Yunnan Baiyao (1g/ml,0.5g/ml,0.25g/ml,0.125g/ml,0.0625g/ml). The negative control group was soaked in sterile saline. Other papers are placed in Petri dishes cultivated with Staphylococcus aureus,Escherichia coli, Pseudomonas aeruginosa, Candida albicans.To evaluate theinhibitory effect of water soluble of Yunnan baiyao on four kinds of bacterias byobserving the size of the inhibition zone to indirectly prove the positive role ofYunnan Baiyao in the process of anastomotic healing.Meanwhile,to observetheinhibitory action ofthe water soluble substances of Yunnan Baiyao for SW480cellsofcolon carcinoma.Part Ⅲ: To study the effect of Water solution of Yunnan Baiyao anti-tumor. Takepre-prepared with different concentrations of the Yunnanbaiyao water solubles,SW480cells were inoculated in96-well plates with200μl,which were set up a controlgroup and4drug experimental groups, with four vice empty.cell culture24hoursafter different concentrations of drugs by adding20μl of the experimental group,placed in a370C incubator to continue to foster, in the cells for24h of MMT20μlobserved the Yunnan Baiyao of different concentrations of water solubles role in cellsurvival rate of cultured cells after24hours.Results:1. The rats were anesthetized satisfactorily,5-0medical absorbable sutureanastomosis of bowel, the distance between theneedlesof all the8-10needleswas0.1cm,there was not leakage by injecting physiological saline into the enteric cavity.The spray coating of the stomaswas uniform. The surgery went well with about30-40min.2. Anastomotic situation:there was a good blood supply and a slight congestionand edema on the stomasfor the postoperative3days. The restitutionwaswell for thepostoperative5days, and a small amount of scar tissues around it were found. A largenumber of scar tissues around the anastomosisand some slight adhesions withmesentery were found for7days. The two drugs are absorbed completely.3. Peritoneal adhesion: there were varying degrees of adhesion in the threegroups. The peritoneal adhesion index of Yunnan Baiyao group was significantly higher thancontrol group(p<0.05) on the5th, andhigher thancontrol group and PNSgroup (p>0.05)on the3rd,7th;The peritoneal adhesion index ofPNS group was higherthancontrol group (p<0.05) on the3rd,5th,7th.4. Bursting pressure of the situation: after three days, Theanastomotic burstingpressure of Yunnan Baiyao group was significantly higher than control group (p<0.05), higher than the PNS group (p>0.05), and the PNS group was also higher thanthe controls(p>0.05); Over time, each group of anastomotic bursting pressure tendedto increase, the Yunnan Baiyao group was significantly higher than the PNS groupandcontrol group (p<0.05)on the5th, PNS group was slightly higher than the controlgroup (p>0.05);The Yunnan Baiyao group and PNS group were significantly higherthan the control group for7days(p <0.05).5. Hydroxyproline content: Over time,the HYP contentof all thegroups increasedslowly. The HYP contentofYunnan Baiyao group were higher than the PNS group andcontrol group(p <0.05)on the3rd,5th,7th;the PNS group was significantly higher thanthe control group (p<0.05)on the3rd,5th;and the PNS group is slightly higher than thecontrol group for7days (p>0.05).6. HE staining: over time, each group of anastomotic inflammationattenuatedgradually.The Inflammatory reaction of Yunnan Baiyao group is moreslight,which was significantly lower than the PNS group and the the control group(p<0.05).7. Masson staining: collagen fibers was colored blue, muscle and cellulose wascolored red,andcell nucleus was black and blue. Anastomotic tissue was shown in asmall amount of collagen fibers, whichwere thin and messy for3days.Over time, thecontent of collagen was more and more, and its orientation showed much moretropism. At the same time, the anastomotic collagen content of Yunnan Baiyao groupwas significantly higher than the control group and PNS group, which was thicker,dense, and the tendency was more pronounced. PNS group also showed a slightadvantagecomparedto the control group.8. The CD31/VEGF Immunohistochemical results: over time, the positiveexpression cellsof CD31/VEGF of all the groups showed a gradually increasing,and Yunnan Baiyao group was more obviousat the early. Over the same period, theexpression rate of Yunnan Baiyao group was significantly bigger than the PNS groupand control group3rd,5th, and the PNS group was higher than the control group. Butthe7th postoperative day, the advantageof Yunnan Baiyao and PNS groupswereslightly weakened.9、The water soluble substances ofYunnan Baiyao were shown to a certaindepressant effect tothe growth of four bacteria for24h, which was the most obvious at1g/ml10、The water soluble substances of Yunnan Baiyao restrained the growth ofSW480cells ina certain extent for24h,andthe IC50was0.27g/ml.Conclusion:1. Yunnan Baiyao or PNS using on the Local has a significant role in promotinggastrointestinal anastomotic healing at the early days, And the effect of Yunnanbaiyaois better than PNS,s2.Yunnan Baiyao water solubles can inhibit the growth of Staphylococcus aureus,Pseudomonas aeruginosa and other bacteria, prevention of anastomotic local infection,thus promoting the healing of the anastomosis.3.The water soluble substances of Yunnan Baiyao restrained the growth ofStaphylococcus aureus,Pseudomonas aeruginosa,andthe otherbacterias, which preventthelocalinfection ofthe anastomosis.4.The water soluble substances of Yunnan Baiyao has a certain inhibitory effectto SW480cells, and has a concentration-dependent.
