Background Mesenchymal stem cells are multipotentdifferentiation potential of non-hematopoietic stem cells, has beenwidely used in stem cell transplantation, tissue engineering and organtransplant immune therapy, especially in Engineered blood vesselsshowed MSC application prospects, but its specific mechanism is notfully understood.Objective To investigate effects of hypoxia on bone marrowmesenchymal stem cells proliferation and adhesion in rats.Method A total20of male or female SD rats, weighing100-120g, were applied in studies. Cells were harvested from femoral bone andtibia of bone marrow cavity. Hematopoietic cell were removed bycell-adhesion method. Expression of CD54,CD29,CD90and CD45 were identified by flow cytometry. Bone marrow mesenchymal stemcells were cultured under atmosphere of1%O2and21%O2respectivelyfor0to7days, Cells growth curve were made by MTT method. Cellsdoubling time were calculated. Cells for Adhesion ability wereclassified into4groups by oxygen concentration at culture andmigration:20%O2culture and20%O2Shear Stress(group A),1%O2culture and1%O2Shear Stress(group B),20%O2culture and1%O2Shear Stress(group C),1%O2culture and20%O2Shear Stress.Migrated cells were counted.Protein levels of Integrinbeta1,HIF-1,VCAM1and ICAM1in MSCs cultured under differentconcentrations of oxygen detected by Western blot.
