Background and objective: Spinal cord injury is the most seriouscomplications of spinal fractures, It may lead to autonomic dysfunction anddifferent degrees of bladder function, sexual dysfunction, urinary tractinfection, abnormal semen quality,etc. clinically, many patients are youngmen, there is an urgent requirement of procreation. because nerve cells cannot regenerate, the treatment of primary lesions after spinal cord injury isstill lack of effective means. foreign studies have shown thatelectroejaculation, penile vibratory stimulatio, percutaneous epididymalsperm aspiratio were used to obtain the patient’s semen, and many patientsby the number of training can penile erection and normal ejaculation.Therefore, abnormal sperm is the main reason for the reduction or loss offertility in patients with SCI. These changes include: the number of bacteriaincreased, reduced sperm count, decreased activity and abnormalchromosomes,etc. the current studyies focused on the urinary tract infection,endocrine disorders, spermatogenic cell apoptosis. in addition to therecovery of spinal cord function, to study new methods of recovery in patients with spermatogenic function is very necessary.To study the expressions of IL-1, TNF-α, SOD and bacterial infectionin male rats’ reproductive system after spinal cord injury, adult male ratswere used to create the animal models of spinal cord injury. it provides atheoretical basis for clinical treatment.Methods:80SD rats were divided into control group(40rats) andexperimental group (40rats) randomly. animal models of spinal cord injurywere established by “Allen” method. both groups of rats were fed for1day,2days,4days,8days, and16days respectively. then8rats’ prostates, testesand epididymises in each group were extirpated for bacterial culture andimmunohistochemistry bySABC to detect the expressions ofproteins(IL-1,TNF-α,SOD) at each time point.Results: Experimental group,40(100%) rats showed flaccid paraplegiaof the hind limbs and no wagging tail activities. there were32(80%) ratswith the bulging of the bladder area after24hours.when the bladder wassqueezed, hematuria is expelled from the urethra and fecal incontinence.eight (20%) rats did not discharge hematuria but there were dysuria andgastrointestinal dysfunction symptoms. the urines of12(30%) rats werecloudy and were sepsis-like after72hours. the hematurias of rats in theexperimental group were disappeared after a week. the control group hadno abnormalities after recovery from anesthesia.The total number of positive samples by bacterial culture in the experimental was46, accounted for22cases of prostate, accounting for20cases of epididymis, its were increased significantly compared with thecontrol group, the difference was significant (P<0.05). testis only4specimens were culture positive, compared with the control group, there wasno significant difference (P>0.05). the numbers of prostates andepididymises’s positive samples in the experimental group were higher thanthe control groups, the difference was significant (P<0.05). less testicularinfection,nosignificant difference compared with the control group(P>0.05);types of bacteria, including E.coli, followed by Enterococcus faecalis, andStaphylococcus aureus.IL-1, TNF-alpha, the most common expression were in testis germ cells,interstitial cells and sperm cells. IL-1positive cells were increased (P<0.05),experimental group and control group for comparison,the number ofpositive cells were in TNF-alpha also increased at each point in time(P<0.05). comparison between the experimental group at the different timepoint: IL-1, TNF-alpha expression were differences (P<0.05). pairwisecomparison, after surgery on the1st and16st the number of positive cellswere the least, IL-1expression was the highest and the difference wassignificantly after surgery on the4th (P <0.05). TNF-α positive cells was theleast after surgery on the1st and was the most after surgery on the4st,compared with other experimental group there was significant difference(P<0.05).SOD is only a small amount of prostate gland cell membrane, the expression of the epididymis main cell membrane and cytoplasm ofspermatogenic cells. SOD Expression,there was no significant differencebetween the experimental group and control group and at different timepoints the experimental group at the same time point.Conclusion: Prostate and epididymis of the rat after spinal cord injuryare easily infected by bacteria(E.coli); inflammatory response wasenhanced in testicular, and the antioxidant activity of reproductive systemmay be very weak.
