To aim of this was to observe emodin-mediated cytotoxicity and theinfluence of emodin on Rad51and ERCC1expressions in non-small celllung cancer (NSCLC). NSCLC were cultured in vitro with emodin at variousconcentrations (0,25,50,75and100μmol/L) for48h and the proliferationinhibition rate of NSCLC was determined by MTT method. Then, NSCLCwere treated with emodin (SK-MES-140μmol/L, A54970μmol/L) or20μmol/LU0126(an ERK inhibitor) for48h, or with various concentrations ofemodin for48h; or with emodin for different durations and the protein andmRNA expressions of ERCC1and Rad51were determined by RT-PCR andWestern blot assay, respectively. Emodin exerted a suppressive effect on theproliferation of NSCLC in a concentration dependent manner. The proteinand mRNAexpressions of ERCC1and Rad51in NSCLC were significantlydecreased following treatment with emodin, which was also in a dose andtime dependent manner. Vacuolar degenerations were observed in A549andSK-MES-1cell lines after emodin treatment by transmission electronmicroscope. Emodin inhibited cell proliferation in NSCLC by downregulation ERCC1and Rad51.
