| Background and Objective:Blood pressure variability(BPV) is a basic physiological characteristics of human beings.It is particularly significant especially in patients with essential hypertension that determines the target organ damage and prognosis as blood pressure levels.The pathogenesis of blood pressure variability has not yet been elucidated.Many studies have recently found a non-selective cation channel on the cell membrane which is canonical transient receptor potential(TRPC).C3subtype(TRPC3) and C6subtype(TRPC6) are closely related to the pathogenesis of hypertension.We build the abnormal blood pressure variability model by sinoaortic denervated(SAD), To investigate the effect of SAD on the expression of TRPC3channel and TRPC6channel in the aorta of spontaneous hypertensive rats(SHR),and the relationship of TRPC3,TRPC6and blood pressure variability.Methods:Twenty male spontaneous hypertensive rats at the age of10weeks were randomly divided into two groups:SAD operation group(n=10)and Sham operation group(n=10).Twenty-four-hour blood pressure and BPV were measured in conscious,unrestrained rats eight weeks after operation.The expression of messenger ribonucleic acid(mRNA) and protein of TRPC3and TRPC6were determined by Reverse transcription polymerase chain reaction (RT-PCR),Western Blot and immunohistochemistry respectively.Results:Compared with SO group,24-hour average systolic blood pressure(SBP) and diastolic blood pressure(DBP) had no significant difference,24-hour systolic blood pressure variability(SBPV) and diastolic blood pressure variability(DBPV) and the expression of mRNA and protein of TRPC3and TRPC6were significantly increased in SAD group (P<0.05)Conclusion:The SBPV and DBPV of SAD group increased,meanwhile, the expression of TRPC3and TRPC6mRNA and protein increased,which implied that the increase of TRPC3and TRPC6were closely associated with elevated BPV,and those might be involved in the regulation of BPV. |
