Background:Pulmonary fibrosis is a chronic, refractory disease. Which damagepulmonary morphology and its function along with the progress, evenleading to organ failure. This disease seriously endangers the patients ’ lifeand health, At the same time it has serious effect on the social and economicdevelopment. So fibrosis treatment has always been the difficulty andhotspot in medical research. Traditional Chinese medicine (TCM) has aunique value in theory and practice on pulmonary fibrosis for liver, kidneyand other organs. Therefore, research and development of new anti-fibrosisdrugs of TCM has the profound social and economic significance.Objective:To evaluate the effect of therapeutics of benefiting qi, expelling bloodstasis and dissipating phlegm on TGF-β1、PAI-1and PPARγ of Rats withPulmonary fibrosis; and to observe lung tissue ultrastructure with thistherapeutics;to study the possible mechanisms that therapeutics of benefiting qi, expelling blood stasis and dissipating phlegm can treatPulmonary fibrosis.Methods:Adopt the model of induce Pulmonary Fibrosis in rats by disposableendotracheal injection of BLM (bleomycin)5mg kg-1d.100rats wererandomly divided into7groups, including model group, normal group, theexpelling blood stasis group, the benefiting qi group, the dissipating phlegmgroup. the hormone group, and the benefiting qi,expelling blood stasis anddissipating phlegm group. Normal control group and model group weretreated with normal saline. Hormone group was given subcutaneousinjection of hydrocortisone while the rest groups were given correspondingmedicines (the expelling blood stasis decoction, the benefiting qi decoction,the dissipating phlegm decoction, and the benefiting qi, expelling bloodstasis and dissipating phlegm decoction) separately for28days after thesemodels were established.2months after such intervention treatment, these7groups of rats were killed to detect the lung tissue expression of PPARgamma through immunohistochemical method, to measure plasma PAI-1levels in each group rats by ELISA, to detect the lung tissue expression ofTGF-β1protein by Western Blot and observe the ultrastructure of the lungtissue through electron microscopy. At the same time, to observe the spirit、activity and weight of all rats before and after molding and drug intervention.Results: Compared with the normal control group rats, the molding rats of allgroups were poor in generally, but improved after the Chinese traditionalmedicine and hormone intervention treatment, and the hormone groupimproved slightly obvious; compared with the rats of model group, plasmaPAI-1levels in the benefiting qi,expelling blood stasis and dissipatingphlegm rats were significantly reduced (P <0.01); Pulmonary tissue of TGFbeta1protein expression decreased significantly; compared with the modelgroup rats, Lung tissue PPAR gamma expression was enhanced intraditional Chinese medicine groups, and most obvious in the benefitingqi,expelling blood stasis and dissipating phlegm group; compared with thenormal control rats; the organization structure in lung with therapeutics ofbenefiting qi, expelling blood stasis and dissipating phlegm was closest tothe normal group.Conclusion:The therapeutics of benefiting qi,expelling blood stasis anddissipating phlegm can significantly inhibit expression of TGF beta1andPAI-1in rats with BLM-induced, and increasing PPAR gamma expression,thus effectively improving the pulmonary fibrosis of BLM-induced rats.
