Objective: Comparative study the differences of distribution frequency between colorectal cancer patients specimens and healthy normal people specimens on three different genotypes TT, TG, GG and rs3809728AA, AT, TT,analysis the association between the two site gene polymorphisms and colon cancer susceptibility. To further explore the relationship between two sites different genotypes and colorectal tumor stage, histological type, differentiation, lymph node metastasis clinical pathological factors,provide basis theoretical for colorectal tumor high risk population screening and early diagnosis.Method: Collect120tube diagnosed with colon cancer f patient’s peripheral blood and120tube normal healthy people peripheral blood.And collect patient group and control group clinical pathological data and health questionnaire, create the database.Extraction genomic DNA from the blood.The polymorphisms of mkk4gene in120colon cancer specimens and120normal people specimens were tested by polymerase chain reaction restriction fragment length polymorphism PCR‐RFLP.Immunohistochemical method was used to detect the expression of mkk4protein. Analysis of the relationship relationship between different genotypes and the tumor site, size, stage, histologic type, degree of differentiation, lymph node metastasis, prognosis clinicopathological parameters.Result: The rs3826392(T/G) site of genotype(T/G+G/G)frequencies in colon cancer patients were significantly lower those in normal people group.(P=0.043,OR=0.576,95%CL=0.337‐0.985). The expression of mkk4protein was markedly increased in tumor tissues with G variation(P<0.001); While rs3809728(A/T) site of T variants show no obvious differences (P=0.142). Rs3826392(T/G) sites of the G allele significantly reduced incidence of lymph node metastasis of probability compared to the T allele (P <0.001, OR=0.218)but showed no significant difference in the degree of differentiation, tumor extent factors (P>0.05). And there is no statistical significancers of3809728(A/T).While rs3809728no significant difference in different genotypes on susceptibility to colon cancer, differentiation degree, scope of tumor, lymph node metastasis factors.Conclution:mkk4gene rs3826392(T/G)site with G variation and the upregulation of the expression of mkk4may be correlated with low incidence of colon cancer,which was a protective factor. And G variation is a protective factor on the lymph node metastasis of colon cancer There were no signifcant correlation betweenrs3809728(A/T) polymorphism and Sporadic colonic cancer.
