Objective: To investigate expression of HA117in bone marrowmononuclear cells (BMMNC) of acute leukemia patients(Als), as well asthe relationship between the HA117gene expression and MDR, Besides,the relationship between HA117gene expression and antigen presentationin acute myeloid leukemia (AML) cells and its clinical significance.Methods: HA117gene expressions in50patients with acute myeloidleukemia and acute lymphocytic leukemia,10patients with Idiopathicthrombocytopenic purpura (ITP) were tested using reverse transcriptasepolymerase chain reaction (RT-PCR) technique. Thirty bone marrowspecimens of AML patients,at different phase, were selected, andimmunophenotypes were assayed by flow cytometry.Results:1. There was no significant difference (P>0.05) in HA117geneexpression between ALL78.57%and AML72.22%. The expressions ofHA117gene in both ALL and AML were significantly higher than inITP patients (P=0.011;0.008<0.05).2. The HA117gene was expressed in75.00%of the initially diagnosedpatients and77.78%of the remission patients and69.23%of thenonremission patients, all of them had higher expression than thepatients with ITP, but there was no significant difference among thethree groups.3. There were47.06%and87.88with HA117gene expressioned innon-resistance group and resistance group respectively, and thedifference between the two groups was obvious.4. HA117had positively correlation with CD11b(OR=0.59,P=0.001<0.05), CD33(OR=0.704, P=0.000<0.05) expression in30patients of AML, and had negative correlation with MPO(OR=-0.611,P=0.001<0.05). Conclusion: HA117had correlation with CD11b, CD33, MPO expressionin AML(except APL), they can be biomarkers for evaluating therapyefficacy in this kind of AL. HA117gene overexpression is significantlyassociated with the clinical multidrug resistance prognosis. It may also beassociated with multidrug resistance of acute leukemia. HA117gene canbe the indicators for investigating clinical multidrug resistance andpredicting prognosis of AL.
