Objective1.To investigate the effect of interscapular brown adipose tissue depletion on bodyfat and blood lipids in C57BJ/6L obese mice induced by high fat diet.2.To investigate the effect of medium chain triglycerides on brown adipose tissue inC57BL/6J obese mice induced by high fat diet.Methods1.22male C57BL/6J mice were divided into2groups, i.e., IBAT depletion andcontrol group. The IBATs were excised under anesthesia in IBAT depletion groupand sham operations were done in the control group. The mice in the2groups werefed high fat diet for16weeks. Body weight, body fat mass, Lee’s index, diet intake,food efficiency ratio and blood lipids were recorded and measured.2. Obese male C57BL/6J mice were obtained by feeding them high fat diet.30obese mice were divided into2groups, and fed high fat diet with MCT or high fatdiet with LCT, respectively. The mice were fed high fat diet containing MCT orLCT for12weeks. Body weight, blood biochemical variables, interscapular brownfat mass, and mRNA expression and protein expression of β3adrenergic receptors,UCP1, PKA, cAMP, ATGL, HSL in brown adipose tissue were recorded andmeasured.Results1.After16weeks, significant increases in body weight, body fat mass, Lee’s index,blood triglyceride levels were observed in IBAT depletion group as compared withcontrol group (P<0.05). No significant differences in diet intake and daily foodenergy intake were noted between IBAT depletion and control groups (P>0.05).2.After12weeks, significant decrease in body weight and body fat mass wasobserved in MCT group as compared with LCT group (P<0.05). Greater increasesin brown fat mass was observed in MCT group than in LCT group(P<0.05).Blood TG, TC, LDL-C in MCT group were decreased significantly, meanwhile bloodHDL-C and ratio of HDL-C/LDL-C were increased significantly. Greater increasesin mRNA expression and protein expression of β3AR, UCP1, cAMP, PKA, HSL,ATGL in BAT were observed in MCT group than in LCT group(P<0.05).Conclusion1.The depletion of IBAT induced increased body weight, body fat mass and bloodtriglyceride in C57BJ/6L mice fed high fat diet. The results suggest that the brownadipose tissue plays a role in preventing the development of obesity.2. Effect of MCT on loss of body weight and body fat might be due to up-regulatingexpression of β3AR and activating cAMP/PKA pathway and enhancing fatmobilization and oxidation in BAT and increasing thermogenesis of BAT.
