Objective To screen the differential expression proteins in the childrenneuroblastoma(NB) by proteomics tools.Methods Three specimens were from the patients diagnosed Ⅳstage NB by biopsy atDepartment of Pediatric Surgery of Chinese PLA General Hospital in Beijing. Anotherthree specimens were acquired from the patients underwenttumor excision. Patients age was1.5years to6.25years (average3.17years). Proteinsfrom children neuroblastoma tissue before and after chemotherapy were separated bytwo dimensional gel electrophoresis.The differentially expressed proteins on2D Gelwere digested with trypsin,and then analyzed by nanoUPLC-electrospray ionizationtandem ms(nanoUPLC-ESI–MS/MS).Results After2-DE,maps about tissues before and after chemotherapy were produced.The7differential protein spots were identified by using the ImageMaster2D software,in which2were up-regulated, including Nm23protein and neuropolypeptide h3,5weredown-regulated in the after chemotherapy, including stathmin1, heat shock protein27,mitochondrial short-chain enoyl-coenzyme A, peroxiredoxin1and peroxiredoxin3.Conclusion The differential expression proteins of children neuroblastoma before andafter chemotherapy were successfully identified by2-DE andnano-UPLC-ESI-MS/MS.These experimental results provide useful clues for thein-depth study of neuroblastoma pathogenesis, early diagnosis and treatment.
