Screen The Specific KSHV Immunoregulatory Genes Of Up-Regulated Expressions Of ARGs

Objective: To screen Specific genes which can activate host AIDS Resistance Genes(ARGs) expression from KSHV immunoregulatory genes.Methods: The eighteen ORFs was obtained by PCR, then it was cloned into eukaryotic expression vector PIRES2-EGFP by TA cloning method. Then transfect those Recombinant vectors into Jurkat cells, and estimate transfection efficiency by fluorescent inversive microscopy after24hours. Use QRT-PCR technique to detect the expression levels of those ARGs (A3G、A3F、CCL5、MX1、MX2) in cells after36hours,and finally screen Specific genes which can activate host ARGs expression from KSHV immunoregulatory genes.Results: Analysising of expression levels of ARGs in Jurkat cells which transfected with recombination carriers,we can screen four specific genes (K4,K6,K13,ORF4) which can activate host ARGs expression from KSHV immunoregulatory genes. k4gene can increase A3G,A3F,CCL5,MX1and MX2of Jurkat cells in2.407、3.086、2.549、2.023and2.671times.K6gene can increase A3G,A3F,CCL5and MX1of Jurkat cells in1.580、2.420、7.367and2.064times.K13gene can increase A3G,A3F,CCL5and MX2of Jurkat cells in2.199.1.518、6.713and21.782times.ORF4gene can increase A3G,A3F,CCL5,MX1and MX2of Jurkat cells in1.843、1.236、4.382、2.772and1.415times.Conclusion: K4, K6, K13, ORF4genes which screened from KSHV immunoregulatory genes can activate AIDS Resistance Genes expression in Jurkat cells.
